| Objective:To explore effects of butylphthalide on pyroptosis-related NLRP3 inflammasome Pathway with cerebral ischemia-reperfusion injury in rats.Methods:Healthy male SD rats were divided into 5 groups: sham operation group,the cerebral ischemia reperfusion group,low dose butylphthalide group,middle dose butylphthalide group and high dose butylphthalide group.Effects of butylphthalide on pyroptosis-related NLRP3 inflammasome Pathway with cerebral ischemia-reperfusion injury in rats were investigated by making the behavioral scores,measuring the cerebral infarction volume with TTC staining,assaying the brain slices with HE staining technology and assessing the expressions of caspase-1、NLRP3、IL-1 with immunohistochemistry.Results:Butylphthalide with 20 mg/kg,40 mg/kg and 80 mg/kg dose could decrease the behavior score,reduce cerebral infarction volume,alleviate the brain damage and depress the expressions of caspase-1、NLRP3、IL-1.But the protective effects of butylphthalide with 80 mg/kg and 40 mg/kg were more effective than that of 20 mg/kg.There was no significant difference between the 80 mg/kg group and the 40 mg/kg group(P<0.01).Conclusion:Butylphthalide can alleviate cerebral ischemia reperfusion injury,which may be related to the supress-regulation of pyroptosis-related NLRP3 inflammasome Pathway. |
