Objective:To investigate the effect of HUCMSCs on Th17/Treg in rats with CCl4 induced acute liver injury.Methods:Thirty SD rats were randomly divided into three groups:blank control group(A)(n=10),model group(B)(n=10),and UCMSCs intervention group(C)(n=10).An acute liver injury model was prepared by intraperitoneal injection of 40%carbon tetrachloride olive oil solution.After 24h,UCMSCs were injected intraperitoneally.Blood was collected from the heart after 48 h.ALT、AST、IL-10 and IL-17 were detected by ELISA.Th17 cells and Treg cells were detected by flow cytometry.Compare the changes and compare liver pathological changes.Results:The levels of AST and ALT in group C were lower than those in group B(P<0.05).The level of IL-17 in group B was higher than that in group A(P<0.05).The level of IL-17in group C was lower than that in group B(P<0.05).The level of IL-10 in group B was higher than that in group A(P<0.05),and the level of IL-10 in group C was higher than that in group B(P<0.05).The level of Th17 cells in group B was higher than that in group A(P<0.05).The level of Treg cells in group B was higher than that in group A(P<0.05),and the level of Treg cells in group C was higher than that in group B(P<0.05).Conclusion:UCMSCs can improve the biochemical parameters of rats with CCl4 induced liver injury and reduce the pathological inflammatory changes of liver.UCMSCs induced the decrease of serum IL-17,the increase of IL-10 and the decrease of Th17/Treg ratio in rats with acute liver injury.Thereby inhibiting excessive immune response,reducing further damage of liver cells,improving local microenvironment to promote regeneration and repair.
