| The Platycodon grondiflonm(Jacq.)A.DC is a traditional Chinese medicine and food in china and many other countries with a long history.The Platycodon grondiflonm(Jacq.)A.DC has multiple pharmacological activities.For examples,it obtained the activities of antitussive,expectant,anti-inflammation and anti-cancers.In the 1990~2005 edition of the Chinese Pharmacopoeia,the total saponin content was not less than 6.0%,and then(2010-2015 edition)was revised to contain platycodin D content of not less than 0.10%.The previous study show that Platycodon grandiflorum has widely distributedand various factors such as provenance(genetic),ecology,and processing methods affect the quality of medicinal materials,and the quality of platycodon from different origins with large difference and also it relationship between total saponin content and saponin D content without significant correlation;There are large differences in antitussive or expectorant effects between samples from different sources,and it has no significant correlation or regularity with the total saponin and platycodin D content in the medicinal materials at the lowest effective dose(0.2g·kg-1),therefore,the quality evaluation and control of Platycodon grandiflorum takes the total saponin or platycodin D as a factor to be further studied;In addition,the method for the identification of platycodon grandiflorum by TLC is vague in the Pharmacopoeia and uses chloroform extensively in the process of extracting and unfolding the medicinal materials.It is of huge significance to further clarify the operation requirements and reduce the amount of chloroform used.In view of these backgrounds,this paper used different sources of platycodon grandiflorum as the research object,carried out a series of research on the quality evaluation and control index of platycodon sinensis and the thin layer identification method of platycodon sinensis,and optimized the quality control method of platycodon grandiflorum.The main methods and results of this paper are as follows:1.The antitussive and expectorant activities of different extracts from Platycodon grandiflorum were studied by ammonia spray method and mouse trachea phenol red excretion method.The components and abundance distribution of saponins in different parts were analyzed by UPLC/Q-TOF-MS/MS.The relativity between the activity of antitussive and expectorant of Platycodon grandiflorum and the components of Platycodon grandiflorum was discussed.The results showed that there was a significant correlation between the antitussive and expectorant activities of Platycodon grandiflorum and the components of Platycodon grandiflorum saponins,and their antitussive activities were closely related to the composition and content distribution of the multi-components of Platycodon grandiflorum saponins.Therefore,the total saponin of Platycodon grandiflorum is still an important factor in the quality control and evaluation of Platycodon grandiflorum.2.The total saponin fingerprints of 31 samples of Platycodon grandiflorum in 10 provinces and cities nationwide were studied by HPLC-UV method,and the similarity of fingerprints was analyzed by the Chinese Traditional Medicine Chromatographic Fingerprint Similarity Evaluation System(2012).The results showed that the similarity of total saponins fingerprints of Platycodon grandiflorum from different origins in China ranged from 0.673 to 0.985,of which 23 samples with similarity higher than 0.9 accounted for 74.19% of the total samples,and 25 samples with similarity above 0.88 accounted for 80.65% of the total samples.The results showed that there were some differences in the total saponins of Platycodon grandiflorum samples from different origins in China,but the similarity of fingerprints was high,which indicated the feasibility of using total saponins of Platycodon grandiflorum as quality control and evaluation index of Platycodon grandiflorum.3.For the determination of content,high performance liquid chromatography(HPLC-UV)method was used to detect the contents of seven major saponins,platycodin D,platycodin D2 and D3,and saponins D and D2.A total of 31 samples from the provinces and municipalities were analyzed,and the total saponin content of the platycodon grandiflorum was analyzed using the 2005 Chinese Pharmacopoeia.Studies have shown that among the 31 samples,the total saponin content of platycodon grandiflorum is more than 6%,93.55%,and the total saponin content is 11.99%.The total content of the seven saponins ranged from 0.2182% to 1.0218% with an average of 0.5096%,revealing that the platycodon grandiflorum samples still contain a large amount of other saponin components.The content of platycodin D is between 0.082% and 0.4961%,with an average of 0.2241%,which is significantly higher than the other six saponin components(the average of the six components is between 0.008% and 0.0751%),but these The content of saponin content was significantly different between samples,and did not show a positive correlation with the total saponin content in the sample.It is proved that platycodin D and total saponins have reasonable rationality as indicators for quality control and evaluation of platycodon grandiflorum.4.The conditions were optimized and the TLC plates,sample preparation methods,sampling and solvent system and heating temperature in the TLC identification method of Platycodon grandiflorum prescribed in the Chinese Pharmacopoeia 2015 edition were investigated.The results showed that good identification results could be achieved under the conditions of using silica gel H plate,using petroleum ether-ethyl acetate eluted system,10 ml sample size and roasting at 105 C for 150 seconds,thus clarifying the requirements of TLC identification terminology and reducing the amount of chloroform. |
