| Objectives To study whether circulating c EPCs、SDF-1 and MMP-9 of acute ischemic stroke in peripheral blood are dynamic and the correlationship of c EPCs、SDF-1 and MMP-9 with the outcome of acute ischemic stroke.Methods Select 92 cases of hospitalized patients with acute ischemic stroke(AIS)within 72 h from June 2015 to December 2015 in our neurology department,71 male patients,and 21 female patients,with an average age of 65 plus or minus 12.The cases with acute ischemic stroke are strictly selected in accordance with the diagnostic criteria set by the the Chinese Medical Association Fourth National Conference on Cerebrovascular Disease.Extract 2ml venous blood from acute ischemic stroke(AIS)patients immediately when they are hospitalized and on 7th day in the morning with ethylenediamine tetraacetic anticoagulant(EDTA-K2),and detect circulating EPCs(CD34+KDR+ cells)by flow cytometry(FCM);stromal cell-derived factor 1(SDF-1),matrix metalloproteinase-9(MMP-9)in the plasma was measured using enzyme linked immunosorbent assay(ELISA);At the same time,functional recovery is assessed in the 90 th day by Modified Rankin score(Modified Rankin Scale,m RS).Twenty outpatients with risk factors controled are taken as control for detecting c EPCs,and the level of SDF-1and MMP-9.Results 1 、 Compared with the risk group,the percentage of c EPCs on the 1th day of hospitalization is lower(p<0.05);compared with the percentage of c EPCs on the 1th day of hospitalization,that of the 7th day from the onset of AIS group has a rising trend,but the difference is not statistically significant(p>0.05).2、Compared with the risk group,SDF-1 expression in patients with AIS on the 1st day and 7th day after hospitalization are both rised,the expression of SDF-1 on the 7th day after hospitalization is significantly higher than that of the 1st day,the differences were statistically significant(p<0.01),;compared with the risk group,MMP-9 expression in patients with AIS group in the pathogenesis of the 1st day and on the 7th day after hospitalization rise(p<0.01),but the difference between the expression of MMP-9 on the 1st day and on the 7th day after hospitalization is not statistically significant(p>0.05).3、Within AIS group,compared with poor prognosis group(m RS>2 points),c EPCs percentage of the 7th day of good prognosis group(m RS less than 2 points)elevates(P<0.05);compared with poor prognosis group(m RS>2 points),SDF-1 expression of good prognosis group(m RS less than 2 points)on the 7th day after hospitalization rises(p<0.05),and SDF-1 expression of good prognosis group on the 7th day is significantly higher than that of the 1st day(p<0.01);compared with poor prognosis group(m RS>2 points),expression of MMP-9 of good prognosis group(m RS less than 2 points)on the 7th day after hospitalization significantly increases(P<0.01),and is significantly higher than that of the 1st day(P<0.01).4、The change rate of SDF-1 correlates with m RS(t=1.211.P=0.044),and that of MMP-9 correlates with m RS(t=3.21.P=0.02)and c EPCs on the 7th day after hospitalization correlates with m RS(t=4.608.P=0.011).Conclusions There is dynamic changes of c EPCs and SDF-1,MMP-9 in peripheral blood of patients with AIS;There is correlationship between m RS score of 90 th day and c EPCs,SDF-1 and MMP-9 in peripheral blood of patients with AIS;c EPCs and SDF-1,MMP-9 in peripheral blood of patients with AIS can be used as predictors of prognosis of AIS. |
