| OBJECTIVE: This time,we conducted an animal experiment to detect the changes of TLR4 expression,IL-10 and 5-HT levels induced by TNBs after treated with the Xiezhuo Jiedu Decoction in UC rats,and compared with the efficacy of western medicine named SASP to explore the Xiezhuo Jiedu Decoction a possible target for the treatment of UC and anti-UC recurrence.METHODS: In this experiment,a rat model of UC was replicated using a TNBs/ethanol mixed solution.100 healthy Wistar male rats were randomly divided into 200-220 g,and 10 rats were randomly selected as the blank control group(NG group).The remaining TU model was induced by TNBs/ethanol mixed solution.Then,they were randomly divided into 5groups,17 in each group,which were model group(MG group),SASP group(SG group),Xiaozhuo Jiedufang low-dose group(ZL group),and phlegm-detoxifying side-dose group(ZM group).The group and the turbid detoxification high-dose group(ZH group)were all treated with intragastric administration.The MG group was given 0.9% saline,and the remaining groups were given SASP and different concentrations of turbidity and detoxification according to the group.Lasts 14 days.At the end of the experiment,the rats were observed.Five rats were sacrificed in the blank group.Eight experimental rats were randomly sacrificed to observe colon tissue and serological markers.The remaining rats were pushed into the TNBs/ethanol mixed solution through their anus.After 48 hours,the general condition of the rats was observed.After the sacrifice,the colon tissue was observed and the serological index was detected.The experiment mainly observed the changes of general condition,pathology,TLR4,IL-10 and 5-HT induced by Xiezhuo Jiedu Decoction in rats after UC rats.The experiment wasperformed with SPSS 22.0 for statistical analysis and was statistically significant at P<0.05,P>0.05 had no statistical difference.Results:1.the general situation:After the first modeling,the rats in each experimental group were significantly different from the NG group.The rats in each experimental group were all improved after treatment.After 2 times of modeling,the symptoms of SG group and ZL group were aggravated.The symptoms of MG group,ZM group and ZH group did not change significantly.The rats in group ZH and ZM were better than ZL group,SG group and MG group,and the latter three groups were similar.2.Disease activity index(DAI)score of rats:The DAI scores of all experimental groups decreased after treatment,P<0.05.After 2 times of modeling,the DAI scores of SG group and ZL group were increased in the experimental group before and after 2 times of modeling,P<0.05;MG group,ZM group and ZH group scores have no significant changes being observed,and P>0.05.3.Colonic Mucosal Injury Index(CMDI)score:The CMDI scores of all experimental groups decreased after treatment,P<0.05.After 2 times of modeling,the CMDI scores of SG group and ZL group were increased compared with the same group before and after modeling,P<0.05;while there were no significant changes of MG group,ZM group and ZH group in the CMDI scores,P>0.05.4.Colon tissue pathology:The pathological conditions of colon tissue were all reduced after treatment in each experimental group.After 2 times of modeling,the colon tissue structure of MG group,ZM group and ZH group did not change significantly.The pathological aggravation of the SG group and the ZL group was similar to that of the MG group.5.Colon tissue TLR4 expression:The TLR4 luminosity values of all experimental groups were decreasedafter treatment,P<0.05.After 2 times of modeling,the TLR4 of SG group and ZL group were higher than those of the same group,and P<0.05.While the TLR4 of MG group,ZM group and ZH group were not seen significantly changes,P>0.05.6.Serum IL-10 status:The IL-10 levels increased after treatment in each experimental group,P< 0.05.After 2 times of modeling,the IL-10 of SG group and ZL group were decreased,P<0.05;there were no significant changes of MG group,ZM group and ZH group in IL-10,and P>0.05.7.Serum 5-HT status:The 5-HT content of each experimental group decreased after treatment,P<0.05.After 2 times of modeling,the 5-HT in SG group and ZL group were increased,P<0.05;while there were no significant changes of MG group,ZM group and ZH group in 5-HT,P>0.05.Conclusions:Xiezhuo Jiedu Decoction has the effect of improving ulcerative colitis in the attack of rats,and has the effect of recurring ulcerative colitis in rats.The anti-ulcerative colitis recurrence by Xingzhuo Jiedu Decoction may be caused by reducting of TLR4 expression,up-regulating IL-10 and down-regulating 5-HT. |
