| Objective:In cell experiments,Ptn gene was overexpressed or silenced in pancreatic cancer cells to achieve differential expression of PTN protein.The effects of differential expression of PTN protein on proliferation and apoptosis of Dorsal Root Ganglion neurons(DRGn)and the growth of DRGn axons were observed.Methods:To search the gene sequence of Ptn in the gene bank(NCBI)and construct the plasmid vector that overexpress or silence Ptn gene,then transfect the constructed plasmid vector of overexpress or silence Ptn gene into MIA PaCa-2 pancreatic cancer cells by liposome method,and to design groups: blank control group,blank plasmid group and experimental group.Detect the expression of Ptn gene in pancreatic cancer cells by qRT-PCR,and detect the expression of PTN protein in pancreatic cancer cells by western blot,to verify the effect of overexpress or silence.PTN proteins overexpress or silence in different groups were extracted and added to DRGn.The proliferation curve of DRGn cells was measured by CCK-8 method,and the effects of differential expression of PTN proteins on proliferation and apoptosis of DRGn cells were analyzed.The effects of differential expression of PTN proteins on the growth of DRGn axons were detected by immunofluorescence.Results:Overexpress or silence of Ptn gene in pancreatic cancer cells,the expression of Ptn gene in experimental group was significantly higher or lower than that in blank control group(p< 0.01);The expression of PTN protein in pancreatic cancer cells of experimental group was significantly higher or lower than that of blank control group(p < 0.01);CCK-8 method was used to measure the cell proliferation curve.Overexpression or under-expression of PTN protein had a significant effect on the proliferation of dorsal root ganglion neurons(p < 0.01);Immunofluorescence observation showed that the length and complexity of DRGn axons in the experimental group were higher than or lower than those in blank control group(p < 0.05).Conclusions:1.Overexpress of Ptn gene can increase the expression of PTN protein in pancreatic cancer cells.2.Silence Ptn gene can reduce the expression of PTN protein in pancreatic cancer cells.3.High express of PTN protein can promote DRGn proliferation and axonal growth,and increase axon complexity.4.Low expression of PTN protein can reduce DRGn proliferation,axonal growth and axonal complexity. |
