Objective:Based on LC-MS metabonomics technology,the endogenous metabolic differentials in spleen,kidney and plasma samples of 16-month-old naturally growing mice were analyzed,and the changes of these marker metabolic differentials in male and female mice aged 3 months and 16 months were compared,as well as the regulatory effects of Liuwei Dihuang Pill on these metabolic markers in mice,so as to reveal the intervention phase of Liuwei Dihuang Pill on aging mice.The biological basis of Guan syndrome.Method:Twenty SPF clean ICR mice,male and female,were randomly divided into model group and Liuwei Dihuang pill group from 3 months old to 16 months old.Each group had 10 female and male mice,and another 10 male and female mice of 3 months old were selected as young control group.The young control group and model group were given normal saline with the same volume ratio,while the Liuwei Dihuang pill group was given Liuwei Dihuang pill.Administration.On the 31st day of administration,blood was collected from the orbital vein of mice to prepare plasma,then neck was severed and sacrificed.The spleen and kidney tissues were taken and the samples were frozen in the carbohydrates had different degrees of changes in the spleen tissue of male rats;27 age-related metabolic markers were identified in the spleen tissue of female rats,which were mainly involved in 11 metabolic pathways,including biological markers affecting amino acid metabolism.The largest number of goods.In the analysis of kidney tissue samples,25 age-related metabolic markers were identified in male rats,which participated in 13 metabolic pathways,mainly in amino acid and lipid metabolism;25 age-related metabolic markers were identified in female rats,which mainly participated in 12 changes in amino acid metabolic pathways.2.Based on the data analysis of age-related metabolic markers in three groups of mice:3-month-old young control group,16-month-old model group and Liuwei Dihuang pill administration group,Liuwei Dihuang pill has a regressive effect on these marker metabolic differentials.In the analysis of spleen tissue samples of male rats,Liuwei Dihuang Pills could regulate 13 metabolic markers,including 11 main pathways,mainly on amino acid metabolism and carbohydrate metabolism.In the analysis of spleen tissue samples of female rats,Liuwei Dihuang Pills could regulate 8 metabolic markers by 5 main pathways,mainly amino acid metabolism and citric acid cycle.In the analysis of kidney tissue samples of male rats,Liuwei Dihuang Pills can regulate 10 metabolic markers.There are 7 main pathways regulating amino acid metabolism and sugar metabolism.In the analysis of kidney tissue samples of female rats,Liuwei Dihuang Pills can regulate 10 metabolic markers,and there are 5 main pathways regulating riboflavin metabolism and carbohydrate metabolism.In the analysis of plasma samples of male and female mice,Liuwei Dihuang Pill can regulate 11 metabolic markers.There are 2 and 4 main pathways of regulation.The metabolic pathway of Liuwei Dihuang Pill on male mice is sugar metabolism,while the metabolic pathway of female mice is concentrated on amino acid metabolism.Conclusion:1.Compared with 3-month-old mice,the endogenous metabolites in spleen,kidney and plasma of 16-month-old mice changed,indicating that natural aging would lead to changes in metabolic status of mice.In the course of natural aging,the metabolic changes of spleen tissue in male rats were concentrated on amino acid and sugar metabolism.The metabolic changes of kidney tissue were mainly amino acid and lipid metabolism,and the metabolic changes of plasma were mainly manifested in sugar metabolism.The metabolic changes of spleen tissue in female rats were concentrated on amino acids,lipids and carbohydrates.The metabolic changes of kidney tissue were mainly amino acids and carbohydrates.The plasma was mainly the metabolic changes of amino acids.2.Liuwei Dihuang Pills can regulate the metabolic changes of spleen,kidney and plasma samples of 16-month-old female and male mice in varying degrees,which proves that Liuwei Dihuang Pills can regulate the metabolic changes of all tissues caused by aging.The intervention of Liuwei Dihuang Pill on spleen and kidney tissues of male rats focused on amino acid and sugar metabolism,and in plasma samples mainly on sugar metabolism.The intervention of Liuwei Dihuang Pill on spleen tissue of female rats focused on amino acid metabolism,mainly on sugar metabolism in kidney tissue and amino acid
