Antelope horn is the horn of Saiga antelope.Buffalo horn is the horn of buffalo.Antelope horn and buffalo horn have good antipyretic effect.They are commonly used in children with high fever and have obvious effect.Although antelope horn and buffalo horn are widely used,their active ingredients are not clear,and their mechanisms need to be explored.In this paper,antipyretic effects and mechanisms of antelope horn and buffalo horn and their extracts were studied,and the components of them were compared,which provided a basis for exploration of keratosis.The experimental results are as follows:1.Using the model of infectious fever induced by yeast injection in rats,we found that antelope horn powder,antelope horn water decoction and buffalo horn water decoction could all decrease the body temperature of feverish rats.The acid and alkali hydrolysates of antelope horn and buffalo horn had no obvious antipyretic effect.Antelope horn powder and buffalo horn decoction can reduce the levels of TNF-α,IL-1β,IL-6,5-HT,cAMP and PGE2 in the serum of febrile rats,and can reduce cAMP and PGE2 in the hypothalamus of febrile rats.Pathological sections showed that antelope horn powder and buffalo horn decoction had protective effects on liver and lung in febrile rats.2.In order to understand the differences of chemical constituents of different extracts,it analyzed the components of antelope horn powder,buffalo horn powder and their different extracts.Infrared spectroscopy showed that protein was the major component in these two horn medicines.The characteristic peaks v(C=O)、β(NH)were about 1660 and 1540 cm-1,which were the strongest or sub-strongest peaks.In addition,two-dimensional derivative spectroscopy can distinguish antelope horn and buffalo horn from 1600 to 800 cm-1,which provides a basis for rapid identification.The protein absorption peaks of antelope horn powder,antelope horn water extract,buffalo horn powder and buffalo horn water extract were higher,while the protein absorption peaks of acid and alkali extracts decreased.Especially for alkali extracts,the amide band I decreased significantly,which was related to the destruction of the original protein structure by acid and alkali.It suggested that the antipyretic effect of antelope horn and buffalo horn and their acid and alkali extracts might be related to the degree of protein degradation.3.LC-MS/MS was used to analyze and identify the protein components of antelope horn powder,buffalo horn powder and their water decoction.16 common proteins were identified in antelope horn powder and buffalo horn powder,23 common proteins from antelope horn powder and its decoction,and 36 common proteins from buffalo horn powder and its decoction.There were seven common proteins in the four samples:Q0VD04,A4FUZ0,F1N362,G3N0V2,F1MMI6,P04272 and F1MUY2.4.Amino acid analyzer was used to determine the contents of 20 amino acids in antelope horn powder,buffalo horn powder and their extracts.The content and proportion of amino acids in them were quite different.The content of various amino acids in antelope horn powder was much higher than that in buffalo horn powder.The total amino acid content of the antelope horn powder is 3.46 mg/g,and it is 0.44 mg/g in buffalo horn powder.The proportion of amino acids in antelope horn powder and its water extract was similar,but the content of them in its acid and alkali hydrolysates increased significantly,and the proportion also changed significantly.Glycine and aspartic acid accounted for 36.7%and 20.5%of the hydrolysates of antelope horn acid,38.0%and 10.3%of the hydrolysates of antelope horn alkali.Buffalo horn powder has the same proportion of amino acids in its water extract,but the proportion of glycine and aspartic acid in acid and alkali extract increases significantly,accounting for 27.7%and 20.3%of the total acid hydrolysate,while glycine and serine acid account for 34.9%and 19.6%of alkali hydrolysate.5.ICP-MS method was used to determine 14 inorganic elements in antelope horn powder,buffalo horn powder and their extracts.The contents and proportion of inorganic elements in antelope horn powder and buffalo horn powder were quite different.The contents of Ca,K,Mg,Na and P in antelope horn powder is 4 times more than those in buffalo horn powder.The contents of Ca and P in antelope horn powder accounted for 66.1%and 29.9%of the total content,which was much higher than other elements.Ca is the most abundant element in buffalo horn powder,accounting for 46%of the total content.The contents of Na,K,P,Zn,Al,Fe and Mg are similar,accounting for 53.5%of the total content.The contents of Ca and P in the water extract of antelope horn is about 35 times less than that of the raw powder,while the contents of Ca,Na,K and P in the water extract of buffalo horn is more than 50 times higher than that of the raw powder.The proportion of inorganic elements in the hydrolysates of antelope horn acid and alkali changed obviously,and the contents of Ca and P decreased significantly.The proportion of inorganic elements in the hydrolysates of buffalo horn acid and alkali also changed.The contents of Ca,P,Mg and K elements in the hydrolysates was lower than that in the buffalo horn water extract with antipyretic effect.
