Parkinson’s disease(PD)is an age-related common progressive neurodegenerative disorder in the elderly.It is characterized by relativelyselective degeneration of dopaminergic neurons in the substantia nigra and form of Lewy bodies(LBs).While the causes of PD remain unknown,multiple epidemiological studies suggest that environmental toxins is the causes of PD.At present,the main drug for clinical treatment is Levodopa,which has large side effects and can’t prevent nerve degeneration.Multiple studies suggest that the treatment of traditional Chinese medicine has small side effects and long-lasting effects.Taohongsiwu decoction is one of the classic Chinese medicines for promoting blood circulation and removing blood stasis.In recent years,it has been found to play a good role in neuroprotection.In this experiment,the lipophilic rotenone treatment of C57BL/6 mice was used to construct the PD model,and investigated the neuroprotective effect of Taohongsiwu decoction on Parkinson’s disease mice.The specific modeling method of this experiment referreds to the way that Chen Naihong’s research group is exposed to environmental contact with rotenone.50 male C57BL/6 PD model mice were randomly divided into5groups,with 10 mice in each group.The control group:during the first 40 days mice were not interfered,40 days after,mice were intragastric administration with water for 9 days(25 mL·kg-1·d-1,once a day);The vehicle group:mice were environment-contact administration of 2%DMSO olive oil,40 days after,mice were intragastric administration with water for 9 days(25 mL·kg-1·d-1,once a day);Model group:mice were environment-contact administration of rotenone(7.5 mg·kg-1·d-1),40 days after,mice were intragastric administration with water for 9 days(25 mL·kg-1·d-1,once a day);The mapodar group:mice were environment-contact administration of rotenone(7.5 mg·kg-1·d-1),40days after,mice were intragastric administration with madopar 120 mg·kg-1·d-1(25 mL·kg-1·d-1)for 9 days(once a day);THSWD group:mice were environment-contact administration of rotenone(7.5 mg·kg-1·d-1),after 40 days,mice were intragastric administration with THSWD 5 g·kg-1·d-1(25 mL·kg-1·d-1)for 9 days(once a day).Mice were intragastric administration for 9 days.The behavioral changes of mice were detected by the mouse rotating rod experiment.The pathological changes of dopaminergic neurons in the brain of mice were observed by HE staining.The number of TH immunopositive cells were detected by Immunohistochemistry.Western blot to detectα-synuclein(α-syn),Cytochrome C(Cyt-C),Bax/bcl-2,and caspase-3 expression level of protein content.The results showed that there was no significant difference in the vehicle group on the rotating rod compared with the normal group(557.90±59.80 vs 564.40±62.30,P>0.05).Compared with the vehicle group,the mice in the model group stayed on the rotating rod for a short time.There was a significant difference(80.00±48.47 s vs 564.40±62.30 s,P<0.05),the cells of the substantia nigra were sparse,the nucleolus was blurred,the cytoplasm was concentrated,the number of TH positive cells was significantly decreased(P<0.05),the expression ofα-syn,caspase-3,Cyt-C was increased,The ratio of and Bax/bcl-2 increased;positive drug and Taohongsiwu decoction can reverse the above changes caused by rotenone administration after intragastric administration.In summary,Taohongsiwu Decoction can inhibit the nerve damage of rotenone-induced Parkinson’s disease model mice,reduce the formation ofα-syn aggregates,down-regulate the ratio of Bax/bcl-2,decrease the expression of caspase-3 and Cyt-C,and increase the survival of DA neurons.
