[Background and Objective] Depression is a common mental disorder,the primary cause of disability in the world,and brings a serious burden to society and families.Chronic stress is the main cause of depression and can lead to neuroinflammation.Brain energy metabolism is one of the most basic characteristics of cell life and plays an important role in regulating cell homeostasis and stress response.The purpose of this study is to investigate whether chronic and unpredictable mild stimulation of(Chronic unpredicted mlid stress,CUMS)can induce hippocampal neuroinflammation and whether the mechanism is related to energy metabolism disorder in hippocampus.[Methods] 1.Male SD rats were subjected to chronic unpredictable mild stimulation of(Chronic unpredicted mlid stress,CUMS)for 28 days to establish an animal model of chronic stress.2.MSD method was used to detect the expression of inflammatory factors in hippocampus.3.LC-MS/MS method was used to detect the changes of energy metabolism in hippocampus.[Results] 1.CUMS can induce the occurrence of Hippocampal Neuroinflammation After SD rats were treated with CUMS for 28 days,the changes of inflammatory factors in hippocampus of rats were detected by MSD.The results show that: Compared with the control group,the expression of IFN-γ,IL-1 β,IL-6,KC/GRO,TNF-α in the hippocampus of the chronic stress group was higher than that of the control group;the expression of anti-inflammatory factor(IL-10,IL-13,IL-4,IL-5)in hippocampus was lower than that in control group.2.CUMS can cause disturbance of energy metabolism in hippocampus In order to clarify the potential mechanism of hippocampal nerve inflammation induced by chronic stress in rats,we used LC-MS/MS method to detect the changes of energy metabolism in hippocampus.The results show that: compared with the control group,the glycolysis increased and the oxidative phosphorylation decreased in the chronic stress group.[Conclusions] Chronic stress can induce neuroinflammation in hippocampus of rats,and its mechanism is related to disturbance of energy metabolism in hippocampus.
