Objective: 1.To study the behavioral characteristics of memory maintenance and extinction in the model of associative learning and memory in the presence of fluoxetine and desipramine.2.Using electrophysiological methods to explore the neural basis of memory maintenance and extinction in the brain barrel cortex in the model of associative learning and memory under the action of fluoxetine.Methods: 1.Behavioral experiments: Establishment of the associative learning and memory model the selected experimental animal was C57 mice at 16 days after birth.The experimental mice were divided into four groups: control group(n=13),fluoxetine group(n=13),desipramine group(n=13)and combination group(n=13).The butyl acetate odor stimulation was performed while performing a mechanical vibration stimulation of 5 Hz on the right side of the mouse using a multi-sensory stimulation simulator.Each group of mice was intraperitoneally injected with the corresponding drug 3 hours before the start of the daily stimulation training.Training interval was 2 hours,30 s for each time,5 times a day for 10 consecutive days.Then,on the 11 th to 17 th day of training,to continue to maintain the corresponding reagent intraperitoneal injection but no longer beard sense of touch and smell combined training.O n the 18 th day,the mice were trained in memory recovery,the corresponding reagents were injected intraperitoneally and the joint training was resumed.At the end of the last training session,the mouse’s whisker’s swing was recorded under the condition that the mouse was stimulated with butyl acetate.2.In this experiment,local field potential signal was recorded in mice of the control group and the fluoxetine group,and the selected recording brain area was the left Barrel Cortex of mice.The mice in the control group were trained on the 10 th day and the 17 th day.The local field potential signal was recorded and the 5 Hz mechanical vibration stimulation was performed on the right beard for 10 seconds.After 20 seconds,the micewere given 10 seconds of butyl acetate smell.The mice on the 10 th and 17 th day of the training in the fluoxetine group were given the whisker tactile stimulation and olfactory stimulation according to the same recording method.The local field potential signals in the Barrel Cortex region were recorded,and the field potentials of the mice in the different groups on the 10 th and 17 th day of training were observed.Results: 1.Behavioral experiment results: The angle and frequency of whisker swing in mice of fluoxetine group,desipramine group and combination group increased with the increase of joint training days,and it was significantly improved compared with the control group.Compared with the peak of the tenth day of joint training,after 7 days of forgetting training,the angle and frequency of whisker swing of the four groups of mice were correspondingly reduced,and the reduction of the control group was the largest with the fluoxetine group,the desipramine group and the combination group.After the forgetting training,joint training was given again.The angle and frequency of the whisker swing of the mice rapidly increased to near the peak level.The fluoxetine group,the desipramine group and the combination group had significant differences compared with the control group.2.Field potential experiment results:(1)On the tenth day of joint training,the electrodes were inserted into the barrel cortex of the control group and the fluoxetine group,and the field potential signal was given when the 5 Hz mechanical vibration stimulation of the whisker was given.The frequency of fluoxetine group signal was higher than that of the control group and there was significant difference.When the odor stimulation of butyl acetate was given,the field potential signal appeared in both groups,and the frequency of fluoxetine group signal was significantly higher than that of the control group.(2)On the seventeenth day of joint training,the field potential signals of the barrel cortex in the control group and the fluoxetine group were recorded.When the beard was stimulated with 5Hz mechanical vibration,the field potential signal appeared in both groups,and the signal frequency of fluoxetine group was significantly higher than that of the control group;when the odor of butyl acetate was stimulated,the field potential signals appeared in both groups,but there was no significant difference.Conclusions: 1.The combined use of fluoxetine and desipramine can enhance the ability of associative learning and memory and can also delay forgetting.2.Fluoxetine has an enhancement effect on the establishment of associative learning and memory,and a delaying effect on extinction.3.Desipramine can enhance the ability of associative learning and memory and delay the extinction of memory.
