| Objective : The pharmacognosy characteristics and pharmacodynamic effects of spermatozoa of different basic kidney were compared to provide evidence for their quality and clinical application.To screen the effective sites of renal sperm quality prostatic hyperplasia to lay a foundation for further study of pharmacodynamics;To explore the chemical composition of renal sperm,and to provide the basis for the establishment of the quality standard of renal sperm,the study of its pharmacological effect and the in-depth study of its chemical composition.Methods:1.To compare the similarities and differences of the two basic renal sperm pharmacognosy by means of morphological identification and microscopic identification,so as to provide a basis for the establishment of quality standards and the distinction between the two.The model of prostatic hyperplasia in rats was induced by subcutaneous testosterone propionate injection,and the sperm of different basal kidneys were given intervention.The urine volume of saline load metabolism,wet weight of prostate,organ coefficient and pathological changes in rats were taken as evaluation indexes to compare the efficacy of the sperm of two basal kidneys.2.Methanol and residual parts were extracted from renal sperm by ultrasonic treatment with methanol.The two extracted parts and the BPH rat model of renal sperm intervention were selected for the first time for the effective parts of renal sperm in the treatment of prostate hyperplasia by taking the wet weight of prostate,organ index and pathological changes as evaluation indexes.3.Since kidney sperm is the pathological product of animals,this experiment adopts the method of GB5009.124 to detect the type and content of amino acids in kidney sperm by amino acid analyzer.By TLC and HPLC,the organic components of bibasic progenitor kidney sperm were compared,which provided reference value for the establishment of quality standard of kidney sperm and the differentiation of bibasic progenitor kidney sperm.Finally,the classification of chemical components in renal sperm was further determined by using super high performance liquid chromatography mass spectrometry combined with Compound Discoverer software,endogenous metabolites and traditional Chinese medicine database,and by comparing the structure of mzCloud,Chemspider and other secondary databases,providing a basis for quality control,pharmacological researchand clinical application of renal sperm.4 、 On the basis of validation of efficacy,combined with 1h-nmr metabolomics technology,serum of rats was analyzed,and major metabolic pathways were further speculated by looking for differential metabolites,laying a foundation for the study of renal sperm mechanism.Results:1.The main distinguishing points for the character identification of bibasal progenitor kidney sperm were described as follows: the diameter of porcine progenitor kidney sperm was 0.1-0.3cm,the cross section was obvious and the texture was crisp.The diameter of bovine kidney sperm was 0.1-2cm,the cross section was not obvious,and the quality was solid.The main differences of its microscopic differentiation are: the porcine origin kidney sperm has more irregular square,rectangular or irregular fragments than the bovine origin kidney sperm;Some crystals of porcine kidney sperm are polarized while most of the crystals of bovine kidney sperm are polarized.In terms of comparison of efficacy,compared with the model group,each kidney sperm group could increase the metabolic urine volume of saline load in rats for 6h(p<0.01),and significantly reduce the wet weight and organ index of the prostate tissue in rats(p< 0.05).The difference was statistically significant,that is,kidney sperm had an improvement effect on prostatic hyperplasia.There was no significant difference in the above test indexes(P > 0.05)between the two groups(P > 0.05),that is,there was no difference in the efficacy of the two groups(P > 0.05).2.Compared with the model group,the wet weight and organ index of the prostate tissues in the alcohol extract group,the kidney sperm group,the qianlikang group and the finacion group were all decreased(P < 0.05),with statistically significant differences.There was no significant difference in the wet weight of prostate and organ index between the residual group and the rats(P > 0.05).The results indicated that alcohol extract was the main part of kidney sperm to improve prostate hyperplasia.3.Compared with the control samples,aspartic acid,threonine,serine,glutamic acid,glycine,alanine,cystine,valine,isoleucine,leucine,tyrosine,phenylalanine,lysine,histidine and arginine were detected in the samples.There was no significant difference in the types of amino acids in the sperm of two basic kidney,but the content of amino acids in the sperm of pig kidney was higher than that of bovine kidney.The result of thin-layeridentification showed that the main spots of bibasic proto-kidney sperm were the same in the heliogram after the silica gel plate was sprayed with 10% sulfuric acid-ethanol,but there was one more blue spot of porcine origin kidney sperm than bovine origin kidney sperm,which could be used as a identification point for bibasic proto-kidney sperm.The HPLC results showed that the chemical composition of the two primitive kidney sperm was basically similar,but the porcine kidney sperm had one more component peak than the bovine kidney sperm at 44 minutes,which was the difference between the two.A total of65 possible chemical constituents,mainly organic acids,were identified from the samples by NMR detection and comparison with secondary databases such as mzCloud and Chemspider.4 、 On the basis of the validation of efficacy,the composition of rat serum was analyzed by combining 1h-nmr metabolomics technology,and 15 different metabolites were found.It was further speculated that the main metabolic pathways were mainly involved in energy metabolism and amino acid metabolism.Conclusion : Although there are great differences in the pharmacognosy studies between different primordial kidney sperm,there is no difference in the efficacy of prostatic hyperplasia.Metabolomics studies have identified metabolic differences in pathological changes and speculated that drug efficacy may be related to energy metabolism and amino acid metabolism.It is preliminarily believed that the effective site of renal sperm in the treatment of prostatic hyperplasia is related to the component of alcohol extract site.The amino acid analyzer detected 15 amino acids in the kidney sperm.The results of TLC showed that the sperms of the two basic progenitor kidneys had the same spots and different components.The organic component peaks of renal spermatozoa were determined by high performance liquid chromatography(HPLC).The chemical composition of renal sperm was further determined by ultrahigh performance liquid chromatography mass spectrometry(uhplc). |
