Objective:To investigate the role of CD73 in the invasion and metastasis of nasopharyngeal carcinoma(NPC).Methods:Ten specimens of patients with nasopharyngeal carcinoma confirmed by pathological section and 10 specimens of normal tissues of nasopharyngeal were collected.Tissue RNA was extracted and QT-PCR was performed.Four nasopharyngeal carcinoma cells 5-8F,6-10B,CNE,CNE-2 and human immortalized nasopharyngeal epithelial cells NP69 were cultured,and RNA and total protein were extracted for QT-PCR and Western Blot experiments,and CD73 high expression was selected.Two nasopharyngeal carcinoma cells were transfected with siRNA to down-regulate the expression of CD73,and the expression of CD73 was detected by RT-PCR.The differeNCe between the blank control group and the transfection group was compared by the proliferation test,the scratch test and the Transwell test.Results:(1)The expression of CD73 in nasopharyngeal carcinoma was higher than that in normal tissues of the nasopharynx.(2)Four nasopharyngeal carcinoma cells 5-8F,6-10B,CNE,CNE-2 and human immortalized nasopharyngeal epithelial cells NP69,QT-PCR and Western Blot test showed that nasopharyngeal carcinoma cell line 5-8F and The expression level of CD73 in CNE-2 was significantly higher than that in human immortalized nasopharyngeal epithelial cells and two other nasopharyngeal carcinoma cells.(3)The QT-PCR results after down-regulation showed that the expression of CD73 in NT5E-siRNA transfection group was significantly lower than that in control-siRNA group.(4)The results of cell proliferation test and scratch test showed that the proliferation ability of NT5E-siRNA transfection group was significantly lower than that of control-siRNA group(P<0.05).(5)Transwell migration and invasion experiments showed that in the NPC cell line,the migration and invasion ability of NT5E-siRNA transfection group was significantly lower than that of control-siRNA group(P<0.05).Conclusion:The expression of CD73 in nasopharyngeal tissues of NPC patients is higher than that of nasopharyngeal normal tissues.Decreasing the expression of CD73 in nasopharyngeal carcinoma cells can significantly reduce the proliferation,invasion and migration of nasopharyngeal carcinoma cells.
