Objective:To study the expression of Sin3 A in gastric adenocarcinoma(gastric cancer)and its relationship with clinicopathological parameters of gastric cancer.Methods:The expression of Sin3A in 225 gastric cancer tissues were detected by immunohistochemical SP method.The relationship between Sin3A expression and clinicopathological factors was analyzed by χ2 test.Kaplan-Meier’s method and COX regression analysis were used to analyze survival,and the log-rank test was used to determine their significance.Results:(1)The expression of Sin3A in gastric cancer(60.00%)was significantly higher than that in normal gastric tissue(8.33%,P<0.05).(2)The positive rate of Sin3A in patients with lymph node metastasis was significantly higher than that without lymph node metastasis of gastric cancer(the positive rates of Sin3A in N2,N1 and NO were 72.22%,64.81%and 48.15%,respectively,P<0.05),The positive rate of Sin3Ain patients of stage III gastric cancer was significantly higher than that of stage Ⅰ/Ⅱ(the positive rates were 67.16%,58.54%and 40.00%,respectively,P<0.05),The positive rate of Sin3A in diffused gastric cancer patients were significantly higher than those in intestinal and mixed(the positive rates were 70.91%,52.17%and 39.13%,P<0.05).The positive rate of Sin3A expression in HER2 positive gastric cancer patients was significantly higher than that in HER2 negative.(the positive rates were 79.17%and 57.71%,P<0.05);(3)(4)The 5-year recurrence rate of Sin3 A positive group was higher than that of negative group(81.48%vs62.22%),the median relapse free survival(RFS)time of Sin3A positive patients was shorter than that of Sin3A negative patients(12 months vs 36 months;χ2=18.267,P<0.05),COX regression analysis shown lymph node metastasis,TNM stage and Sin3A positive expression are independent factors for RFS.(5)The 5-year mortality rate of Sin3 A positive group was higher than that in the negative group(80.00%vs 54.44%),the median overall survival(OS)time of Sin3A positive patients was shorter than that of Sin3A negative patients(19 months vs 49 months;χ2=24.104,P<0.05),COX regression analysis shown lymph node metastasis,TNM stage,HER2 positive expression,and Sin3A positive expression are independent factors for OS.Conclusion:(1)The expression of Sin3 A protein in gastric cancer can be used as an index of gastric cancer malignancy;the abnormal expression of Sin3A is related to the development of gastric cancer;(2)The expression of Sin3 A in gastric cancer is associated with lymph node metastasis,TNM stage,diffused subtype and HER2 positive expression;(3)The positive expression of Sin3A in gastric cancer is associated with poor prognosis;(4)The Sin3 A is a potent biomarker and potentialtherapeutic target for gastric cancer. |
