Objective: The purpose of this study was to investigate the effect of heat treatment on proliferation migration and the expression of miR-23a-3p in gastric cancer BGC-823 cells.Methods: When Human gastric cancer BGC-823 cells grown in log phase,some cells were used to be heat-treated(treated at 43℃for 1 hour),and the other cells cultured at room temperature(37℃)were used as control.After the cells were treated,the cells were cultured under normal temperature conditions.The proliferation of cells in each group was detected by MTT at 0h,24 h,48h and 72 h after treatment.After 24 h,the migration of cells in each group was detected by Transwell chamber and the relative expression of miR-23a-3p was detected by qRT-PCR.Results: 1.The results of MTT assay showed that the OD values at 0 h,24 h,48 h and 72 h after treatment in the normal temperature group were 0.5224±0.0023,0.7456±0.0022,1.0425±0.0019 and 1.2971±0.0021;OD values at 0 h,24 h,48 h and 72 h were 0.5181±0.0022,0.7027 ±0.0060,0.9423 ± 0.0048 and 1.1666 ± 0.0041 in the heat treatment group.There was no significant difference in cell proliferation activity between the two groups after heat treatment at 0h.After 24 h,48 h and 72 h,the proliferation activity of the heat-treated group was lower than that of the normal temperature group,and the difference was statistically significant(P<0.05).2.The results of Transwell chamber migration experiment showed that the number of migrated cells was 157.44±3.02 after treatment in the normal temperature group and the number of migrated cells was 86.67±2.18 at 24 h after treatment in the heat treatment group.The migration ability of gastric cancer cell line BGC-823 in the heat-treated group was lower than that in the normal temperature group,and the difference was statistically significant(P<0.05).3.The results of qRT-PCR showed that the expression of miR-23a-3p in gastric cancer cell line BGC-823 was significantly down-regulated in the heat-treated group compared with the normal temperature group(P<0.01).Conclusion: The proliferation and migration ability were decreased and the expression miR-23a-3p was down-regulated of gastric cancer BGC-823 cells after they were treated at 43 ℃.
