| Objective:The effects of intra-articular injection of sodium hyaluronate on articular cartilage and subchondral bone in osteoarthritis of ovariectomized rats were investigated by establishing an in vivo model of OA combined with osteoporosis.Methods:Thirty female SD rats aged 3 months were randomly divided into 3 groups(10 rats in each group):Sham-operated group,Bilateral anterior cruciate ligament transection in ovariectomised rats(OVX-ACLT group),Bilateral anterior cruciate ligament transection in ovariectomised rats with intraarticular injection of sodium hyaluronate(the treatment group).The sham-operation group without any treatment,but in OVX-ACLT group and treatment group with ovariectomized and transected anterior cruciate ligament of knee joint.Two weeks after OVX-ACLT,rats in the treatment group received intraarticular injection of sodium hyaluronate 50μg(0.05cc)once a week for 12 weeks.Rats in the OVX-ACLT group were injected with normal saline.The rats were sacrificed after 12 weeks of treatment.The CTX-I,TRACP5b,BALP levels were detected in blood,and Mankin scores of articular cartilage and Micro-CT bone mass analysis of subchondral bone were also evaluated.Results:(1)The content of CTX-I,TRACP5b,BALP in blood of OVX-ACLT group(43.68±22.25ng/ml,522.84±71.51pg/L,2.17±0.29μg/L)was significantly higher than that of sham operation group(18.21±8.06ng/ml,427.61±33.55pg/L,1.83±0.30μg/L),The difference is statistically significant(P<0.01,P<0.05,P<0.05),It indicates that osteoporosis exists in OVX-ACLT group,while that of treatment group(36.53±10.16ng/ml,484.00±51.37pg/L,1.92±0.26μg/L)was significantly lower than that of OVX-ACLT group(43.68±22.25ng/ml,522.84±71.51pg/L,2.17±0.29μg/L),The difference is statistically significant(P<0.05,P<0.05,P<0.05),This indicates that the treatment group inhibited the loss of subchondral bone mass.(2)Comparison of Mankin scores among three groups:the scores of the OVX-ACLT group(8.89±1.67)was significantly higher than that of the sham operation group(0.96±0.71),The difference is st`atistically significant(P<0.01),It indicates that the cartilage damage in OVX-ACLT group is serious,and the scores of the treatment group(2.56±1.42)was significantly lower than that of the OVX-ACLT group(8.89±1.67),The difference is statistically significant(P<0.01),This indicates that the treatment group delayed the progressive damage of cartilage.(3)The bone volume fraction(25.2451±2.6879%)and trabecular bone numbers(3.3394±0.4663mm-1)in OVX-ACLT group were significantly lower than those in sham-operated group(45.1209±3.2243%,5.5016±0.4170m m-1),The difference is statistically significant(P<0.01,P<0.01),but the degree of bone trabecular separation(208.6545±24.6400μm)was significantly higher than that in sham-operated group(100.0690±7.0491μm),The difference is statistically significant(P<0.01),the trabecular thickness was not statistically different between the two groups(P>0.05),This indicates the presence of osteoporosis in the subchondral bone of the OVX-ACLT group.The bone volume fraction(43.1300±4.3086%)and trabecular bone number(4.7820±0.4009mm-1)in treatment group were significantly higher than those in OVX-ACLT group(25.2451±2.6879%,3.3394±0.4663mm-1),The difference is statistically significant(P<0.01,P<0.01),but the degree of bone trabecular separation(121.4571±19.6099μm)was significantly lower than that in OVX-ACLT group(208.6545±24.6400μm),The difference is statistically significant(P<0.01),the trabecular thickness was not statistically difference between the two groups(P>0.05),This indicates that the strength of the subchondral bone in the treatment group is improved.Conclusion:Injection of sodium hyaluronate into articular cavity can improve the strength of subchondral bone and delay the degeneration of articular cartilage in ovariectomized rats. |
