Objective micro RNA-33 can inhibit ABCA1,increase cell lipid storage and promote the formation of atherosclerotic plaque.Cyclic RNA can be used as mi RNA sponge to inhibit the expression of mi-RNA.hsacirc0076981 and mi R-33 a have binding sites.The purpose of this study is to investigate the effects of hsacirc0076981 on mi R-33 a,ABCA1 and intracellular lipid accumulation.Methods THP1 cells were induced and differentiated into macrophages by phorbol ester(PMA).The cells were divided into blank control group and experimental group.The blank control group was macrophages.ox-LDL was first added to the experimental group to promote lipid accumulation of cells to form foam cells.Each experimental group was transfected with si RNA or adenovirus overexpression vector as follows.Ox-LDL is a pure foam cell control group: macrophages further react with ox-LDL for 48 hours to form foam cells.Ox-LDL+adenovirus NC was negative control group: blank vector transfected foam cells.Ox-ldl+adenoviral hsacirc0076981 is the overexpression group: hsacirc0076981 adenovirus vector transfects foam cells.Ox-LDL+hsacirc0076981 si RNA is the interference group: hsacirc0076981 si RNA vector transfects foam cells.6.Ox-LDL+hsacirc0076981 Scramble group: hsacirc0076981 scramble vector transfected foam cells.Real-time fluorescence quantitative PCR was used to detect the expression levels of hsacirc0076981,mi R-33 a and ABCA1 m RNA.Western blot detected ABCA1 protein level.Oil red O staining was used to observe cell lipid droplets.Results q PCR and WB analysis showed that Hsacirc0076981 could inhibit mi R-33 a and up-regulate ABCA1 expression.Oil red O staining was used to observe hsacirc0076981 to observe lipid accumulation in basal cells.Conclusion hsacirc0076981 can inhibit mi R-33 a,up-regulate the expression of ABCA1,and reduce intracellular lipid accumulation.
