In this study,Wikstroemia chamaedaphne Meisn.and Diaporthe phaseolorum FS431 were chosen to research their chemical constituents.The chemical constituents were consecutively separated by silica gel,Sephadex LH-20 colum chromatography,ODS chromatography,semi-preparative HPLC and preparative HPLC.The compounds were further elucidated by their physicochemical properties and extensive spectroscopic analysis(1D/2D-NMR,MS,IR,UV,ECD).A total of 40 compounds were obtained from the two subjects,of which 21-23 were new compounds."Huang yuan hua",is the bud of Wikstroemia chamaedaphne Meisn..Twenty compounds were isolated from on the methanol extract of the dry buds of Wikstroemia chamaedaphne Meisn.These compounds were identified as syringaresinol(1),5,7-dihydroxychromone(2),5-hydroxy-7-methoxychromone(3),luteolin(4),pinoresinol(5),caffeic acid methyl ester(6),lariciresinol(7),eudesmin(8),sesamin(9),eriodicty-ol-7,3’-dimethyl ether(10),piperitol(11),forsythialan A(12),pinoresinol-4-O-β-D-glucopyranoside(13),torilin(14),blumenol B(15),eriodictyol(16),naringenin(17),shonanin(18),4-hydroxyl ethyl cinnamate(19),caffeic acid ethyl ester(20).Compounds 2,3,6,10,11,12,14,15,16,18,19,20 are isolated from Wikstroemia Endl.for the first time.Compounds 2,3,6,9,10,11,12,14,15,16,17,18,19,20 are isolated from this plant for the first time.The effects of inhabitation of some compounds on the proliferation of human tumor cell lines were detected by CCK8 method.In this experiment,7 human tumor cell lines were randomly selected:A549(human lung cancer cells),HepG2(human hepatocellular carcinoma cells),PC-3,DU145(human prostate cancer),7860(human renal clear cell carcinoma),BXPC3(human pancreatic cancer)and MDA-MB-231(human breast cancer)proliferation inhibition.The results showed that only caffeic acid methyl ester showed significant anti-tumor effects.In order to adapt to the high salt,high pressure,low temperature and low nutrition living environment,marine microorganisms could produce a large number of bioactive natural products by forming unique metabolic modes.Some active compounds which were derived from marine microorganisms have been used in clinical treatment.As an important part of marine microbes,marine fungi have been discovered many novel and active chemical structures in their secondary metabolites,which are important sources for the discovery of medicinal lead compounds.The marine fungus in this paper was taken from the sediments of the Indian Ocean and identified as Diaporthe phaseolorum FS431.The strain was developed to large-scale solid fermentation.Twenty compounds were totally isolated and identified from the rice medium:three new compounds 21-23.Some compounds were selected to test a-glucosidase inhibitory activity with PNPG method,antioxidant activity test with DDPH method,and tyrosinase activity with L-dopa method.The compounds were tested their anti-cytotoxic activity against hepatoma cells(HepG-2),breast cancer cells(MCF-7),glioma cells(SF-268)and human lung cancer cells(A549)with SRB method.Inhibition rates greater than 50%were further assayed for IC50 values.The results showed that only compound 31 had inhibitory activity against the above four cancer cells,and the IC50 values were 2.55,2.60,37.96,and 4.64 μg/ml,respectively.Compound 32 had inhibitory activity against the above four cancer cells,the IC50 values were 49.58,48.08,45.26 and 38.64 μg/ml,respectively. |
