Objective : this experiment by acupuncture antihypertensive treatment of spontaneous hypertension rats(SHR)and observe its effect on blood pressure,and detect the Ang Ⅱ,ACE and the content of ET 1 and AT1 R and Profilin-1 in the positive expression of aorta,observe thoracic aorta tissue changes in the morphology,investigate the antihypertensive mechanism of acupuncture step-down party and its intervention effect on vascular damage and their differences,for the prevention and cure of acupuncture prescription of primary hypertension and improve the target organ damage provide some experimental basis and new way of thinking direction,thus to provide a certain experimental study acupoints compatibility mechanism and theory.Methods : thirty SHR rats at 9 to 10 weeks were randomly divided into model group,acupuncture group and losartan group.Another 10 week-old WKY rats were used as blank controls.The blank group and the model group were treated the same way without any treatment.The acupuncture group received treatment between 14:00 and 16:00 daily with acupuncture for 5 days,rest for 2 days and treatment for 8 weeks.Rats in the losartan group were given 50mg/kg and 1ml/100 g daily by gavage,and the treatment cycle and time were the same as those in the acupuncture group.The blood pressure level was observed every two weeks,and the thoracic aortic tissue was taken after 8 weeks of treatment for index detection.Histomorphological changes were observed by HE and Masson staining.The Ang2,ACE and et-1 protein contents in serum were detected by ELISA method,and the profilin-1 and AT1 R positive expression contents in aortic tissue were detected by immunohistochemical method,so as to observe the efficacy of acupuncture antihypertensive prescription in reducing blood pressure and improving vascular remodeling and the possible mechanism.Results:1.Changes of systolic blood pressure of rats in each group at different timesDuring the 8-week treatment,the blood pressure of the model group increased compared with the blank group(P < 0.01).After 2 weeks of acupuncture,the blood pressure of the acupuncture group and the western medicine group was significantly lower than that of the model group(P < 0.05,P < 0.01),and there was no significant difference between the treatment groups(P >0.05).After acupuncture for 4 weeks,the blood pressure of the acupuncture group and the western medicine group was significantly lower than that of the model group(P <0.05,P < 0.01),while the blood pressure of the western medicine group was higher than that of the acupuncture group(P <0.05).After 6 weeks of acupuncture,the blood pressure of the acupuncture group and the western medicine group decreased(P <0.05,P < 0.01),and the blood pressure of the western medicine group was still higher than that of the acupuncture group(P < 0.05).After 8 weeks of acupuncture treatment,the blood pressure of the acupuncture group was lower than that of the model group(P < 0.05),and there were significant differences between the western medicine group and the model group(P < 0.05,P < 0.01).2.Comparison of serum levels of active factors in rats of each groupThe contents of AngⅡ and et-1 in the model group were compared with those in the blank group,and the differences were statistically significant(P < 0.01).After 8 weeks of intervention,the differences between the acupuncture group and losartan group were statistically significant(P < 0.05 or P < 0.01)compared with the model group.Comparison of Ang Ⅱ contents between acupuncture group and losartan group shows that the decline level of losartan group is more significant(P < 0.05 or P < 0.01).ACE content in the model group was significantly different from that in the blank group(P< 0.01).After 8 weeks of intervention,the difference between the acupuncture group and losartan group was statistically significant(P < 0.01).By comparing the ACE content of acupuncture group and losartan group,it can be seen that the declining level of losartan group is significantly lower than that of acupuncture group,and the difference is statistically significant(p>0.05).3.Morphological comparison and analysis of rat aortic remodeling in each groupBy comparing the aortic media thickness(MT)and the ratio of media to inner diameter(MT/LD)of rats in each group,it can be seen that the difference between the model group and the blank group was statistically significant(p<0.01).Compared with the model group,the values of MT and MT/LD in the acupuncture group and losartan group decreased significantly,and the differences were statistically significant(P < 0.05 or P < 0.01).In comparison with the acupuncture group,there was a significant difference in the reduction of vascular damage in the losartan group(P < 0.05).4.Observation and results of Masson staining morphology of aortaThe aorta collagen fibers of SHR rats in the model group accounted for a larger proportion of the total area,and the mesa was significantly thickened,elastic fibers were decreased,broken and disorderly arranged,which was significantly different from the blank group(P< 0.01).Compared with the model group,the situation of acupuncture group and losartan group was significantly improved(P < 0.05 or P < 0.01),the deposition of collagen fibers in the media was significantly reduced,and the elastic fibers were orderly arranged and distributed evenly.Compared with the acupuncture group,there was a significant difference in the reduction of vascular remodeling in the losartan group(P < 0.05).5.Expression of AT1 R and profiin-1 in aorta of each group was observed by IHCImmunohistochemical test showed that the difference between the model group and the blank group was statistically significant(P < 0.01).After 8 weeks of intervention,the positive expressions in the acupuncture group and losartan group were significantly reduced compared with the model group(P < 0.01).The difference between acupuncture group and losartan group was significant(P < 0.01)Conclusion:1.Acupuncture with formula which composed of "renying point","quchi point" and "zisanli point" on SHR have actually antihepertentive effect.2.Acupuncture of "antihypertensive prescription" can reduce the contents of ACE,Ang Ⅱ and et-1 in SHR serum,reduce the expression of AT1 R and profilin-1 in SHR aorta,reduce the damage to vascular structure caused by hypertension and delay the process of vascular remodeling by inhibiting the activity of local RAAS.3.The mechanism of acupuncture in reducing blood pressure may be related to the regulation of local RAAS imbalance and the improvement of vascular remodeling.
