Objective 1 To explore the distribution of TCM syndromes in patients with IPF.2 To investigate the effect of supplementing qi,nourishing yin and promoting blood circulation on VEGF,PI3 K,AKT,HIF-1 α protein in rats with pulmonary fibrosis.Methods: 1Investigation of clinical syndrome: consult and refer to the relevant documents of IPF,complete the establishment of TCM syndrome questionnaire,consult the general data of patients and Chinese medicine diagnosis information through questionnaire form.The distribution of TCM syndromes in patients with IPF was analyzed.2 Animal experiment: 128 male SD rats(body weight 260 ±20g)were randomly divided into four groups: blank group,model group,Shenqi Cordyceps group and prednisone group,with 32 rats in each group.After 3 days of adaptive feeding,the rats in the blank group were treated with intratracheal injection of normal saline for sham operation.The other three groups were treated with bleomycin to make pulmonary fibrosis model,and the rats in the Shenqi Cordyceps group and prednisone group were gavage with the corresponding drugs.The blank group and model group were given intragastric administration of normal saline for 28 days.All groups were taken from the 7th,14 th,21st and 28 th day of drug intervention,respectively.The samples were stained with HE,Masson,and the samples were stained with Western.Blot method was used to detect the expression of VEGF,PI3 K,AKT,HIF-1 α protein in lung tissue of each group.Results: 1 Syndromes investigation The syndrome distribution of 64 patients met the criteria: deficiency of both qi and yin(29.69%,17.19%),phlegm-heat obstruction of lung(23.44%),deficiency of lung and kidney(15.63%),syndrome of qi-yin deficiency(29.69%),syndrome of stagnation of phlegm-heat(23.44%),deficiency of lung and kidney(15.63%).Syndrome of phlegm-dampness obstructing lung(12.50%),deficiency of lung qi(10.93%),syndrome of yin deficiency and internal heat(7.81%).2.1 HE staining In the blank group,the alveolar structure was normal,the alveolar cavity was clear,there was no exudate,there was no obvious thickening of alveolar septum and no collagen deposition.On the 7th day of the model group,more inflammatory cell infiltration was seen in the alveolar cavity,mainly neutrophils,macrophages and lymphocytes,the alveolar space was wider than the blank group,and the partial interval was broken.On the 14 th day,a large amount of inflammatory cell infiltration was seen in the alveolar space,and the alveolar space was widened at the 7th day.In the lung,the fibroblasts were seen to be hyperplastic,and a small amount of collagen was deposited.There were more new blood vessels and some of the pulmonary vessels were occluded.On day 21,the infiltration of inflammatory cells in the alveoli was reduced on day 14,and the deposition of collagen was more severe.On day 28,alveolar structure disorders,fusion,alveolar space were further widened and were large In the amount of collagen deposition,the inflammatory cells were significantly reduced,and there was still focal lymphocyte proliferation.At the same time,the performance trend of Shenqi Cordyceps group and prednisone group was similar to that of model group,the degree of Shenqi Cordyceps group was slightly lighter than that of prednisone group,and the degree of both groups was lighter than that of model group.2.2 Masson staining In blank group,there was no obvious abnormality in the wall of trachea and around the wall of blood vessel.On the 7th day in the model group,obvious thin blue collagen fibers were observed around the airway wall and vessel wall.On the 14 th day,the blue collagen fibers increased and the color increased,mainly located in the lung stroma.On the 28 th day,a large number of dense blue collagen fibers were found in the lung interstitial,which were distributed as bundle strips or large slices,showing obvious pulmonary fibrosis.At the same time,the performance trend of Shenqi Cordyceps group and prednisone group was similar to that of model group,the degree of Shenqi Cordyceps group was slightly lighter than that of prednisone group,and the degree of both groups was lighter than that of model group.2.3 VEGF,PI3 K,AKT,HIF-1 α protein expression On the 7th,14 th,21st and 28 th day,the protein expression of VEGF,PI3 K,AKT,HIF-1 α in the model group was higher than that in the blank group,and there was significant difference between the two groups(P < 0 05).The expression of VEGF,PI3 K,AKT,in the Shenqi Cordyceps group and prednisone group was significantly higher than that in the control group.The expression of HIF-1 α protein in Shenqi Cordyceps group was similar to that in prednisone group,but there was no significant difference between the two groups(P > 0 05).The expression of VEGF,PI3 K,AKT,HIF-1 α protein in Shenqi Cordyceps group was similar to that in prednisone group(P< 0 05).Blank group 4 Tim There was no significant difference in the expression of VEGF,PI3 K,AKT,HIF-1 α protein between the two groups(P > 0 05).Conclusion 1 Qi-yin deficiency and stasis syndrome is a common syndrome type in IPF patients.2.The Chinese caterpillar fungus can inhibit the expression of VEGF protein in the rats with pulmonary fibrosis,thus inhibiting the angiogenesis.3.Supplementing qi,nourishing yin and activating blood circulation could inhibit pulmonary fibrosis to some extent by inhibiting the PI3K/AKT/HIF-1 α signal of pulmonary fibrosis rats to interfere with angiogenesis. |
