Objective:(1)The memory performance of VD Rats was tested by water maze experiment,and the effect of Huayu Tongluo moxibustion on memory ability of VD rats was studied.(2)By using immunohistochemical and immune imprinting to detect the expression of specific proteins in the process of proliferation,migration and differentiation of NSCS,as well as CD34 detection of vascular density around newborn neurons,the effects of Huayu moxibustion on "neurogenesis" and "angiogenesis" were studied.Method:Experiment one: The use of clean grade adult male Wistar rats,through the water Maze experiment to screen it,the reaction is too sensitive or dull rats removed,the remaining rats randomly selected sham surgery group.Using the improved 2?VO method,the VD animal model was reproduced in the screened rats,and the VD rat model was identified by water Maze experiment.In this paper,36 were randomly selected as the effect of Huayu Tongluo moxibustion on spatial memory ability of VD rats,and then these VD rats were randomly divided into moxibustion group,Western Medicine Group,model group,12 each,randomly selected 12 fake operation group as the control.Experimental animals in the mold after 7 days to start treatment,moxibustion group VD Rats to choose "dazhui","baihui","shenting" Acupoint,each acupoint moxibustion 1 times a day,each 20 min,7 days for a course of treatment,a total of 3 courses,treatment between continuous uninterrupted;Western medicine group to rat Western medicine(rat nerve growth factor)peritoneal injection,experimental basis The equivalent dose ratio table of human and animal "determines the dosage of rats,the duration of treatment and the course of therapy with moxibustion Group;The rats in the model group were only fixed in the mouse plate,no treatment,treatment time and course with Moxibustion group;the false modelgroup was not treated.After 3 courses,the spatial memory ability of rats was tested by water labyrinth experiment,and the results were compared between groups and groups,and the effect of Huayu Tongluo on spatial memory ability of VD rats was studied.Experiment II: The method of copying VD model was studied by randomly selecting 108 VD rats from successful animals to study the effects of moxibustion on "nerve freshmen" and "angiogenesis",and the VD rats were randomly divided into moxibustion group,Western Medicine Group and model group,each 36,each group was divided into three sub?groups(one course,two course,three course of treatment,each subgroup 12),and randomly selected false model group 36 control,sham Operation Group is also divided into three sub?groups.The treatment method of animals is the same as experiment one.The rats were executed 24 h at the end of the corresponding course of treatment,and the brain tissue was taken.Through immunohistochemical and Western blot to detect the proliferation of NSCs in the hippocampus of VD rats,the expression of specific proteins during differentiation and the density of blood vessels around newborn neurons,the proliferation and differentiation of NSCs and the density of blood vessels around newborn neurons were studied,and the effects of Huayu Tongluo moxibustion on "neurogenesis" and "The effects of angiogenesis."All experimental data are processed using SPSS23.0,and the measurement data is represented by the mean ±standard deviation,it was statistically significant(P<0.05).There is no statistically significant use of(P<0.05).Result:(1)Water labyrinth Experimental results: Compared with the sham Operation Group,the fifth day of the model group Rat Water Maze Experiment incubation period is extended,the number of crossing platforms decreases,the proportion of time in the quadrant of the platform is reduced,compared with the model group,moxibustion Group and Western Medicine group of rats to avoid shortening the incubation period,the number of crossing platforms increased,the The results were statistically significant.(2)The effects of Huayu Tongluo moxibustion on neurogenesis and angiogenesis in hippocampus of rats: Western medicine,moxibustion group compared with the model group,the expression of neun cells increased significantly(P<0.01),in the same treatment period,the expression of neun cells in Moxibustion group was better than that of Western Medicine Group(P<0.05).Compared with VD model group,the map expression of Western medicine and moxibustion Group increased(P<0.05),compared with VD model group,the AQP4 expression of western medicine and moxibustion Group decreased(P<0.05),which showed that after corresponding treatment,moxibustion and Western medicine were treated with VD.In the model rats,the nerve repair in the hippocampus and the differentiation of neural stem cells were well promoted.Compared with VD model group,the expression of CD34 in Western medicine and moxibustion Group increased(P<0.05),which showed that after treatment,moxibustion and Western medicine could increase microvascular density in ischemic hippocampus of VD model rats,thus promoting angiogenesis,and moxibustion obtained the curative effect of approaching Western medicine.Conclusion(1)The spatial memory ability of VD rats can be improved by Huayu Tongluo moxibustion.(2)Huayu Tongluo moxibustion can improve the expression of Neun,MAP and AQP4 in the hippocampus of VD rats,promote the proliferation and differentiation of neural stem cells after cerebral ischemia,and contribute to the repair of nerve function.At the same time,it can improve the expression of CD34 in ischemic hippocampus of VD rats,increase microvascular density and promote angiogenesis.
