| Objective:1.To investigate the level of miR-21-5p in the serum of intracerebral hemorrhage(ICH)patients,and its correlation with hematoma volume,perihematomal edema volume and prognosis.2.To predict the effect and potential mechanism of miR-21-5p on perifocal inflammatory response after ICH via bioinformatics.Method:20 cases of patients with ICH in the 1,2,3 and 7 days after onset and 10 cases of normal control’s serum were collected,and the expression level of miR-21-5p in serum of each sample was detected by qRT-PCR.The correlation between hematoma volume,perihematomal edema volume,NIHSS score at the time of admission and the mRS score at 3 months of follow-up patients were analyzed.We next performed a bioinformatic analysis to seek the molecular targets of miR-21-5p,which was explored by Go enrichment,KEGG pathway analysis and PPI network.Results:1.miR-21-5p levels were elevated in ICH patients,as confirmed by qRT-PCR.Compared with healthy controls,miR-21-5p expression levels in patients with ICH differed significantly on day 1,2,3,7 is 3.731±0.954,2.665±0.747,2.575±0.707,2.682±0.866(P<0.05).2.The expression of miR-21-5p was significantly correlated with hematoma volume on day 1 and day 2-3(R2day1=0.725,P<0.01;R2day2-3=0.643,P<0.01),but appeared poor correlation on day3-7,(R2day3-7=0.436,P<0.05).The expression of miR-21-5p was correlated with perihematomal edema volume on day 1,day 2-3 and day 3-7(R2day1=0.601,P<0.01;R2day2-3=0.667,P<0.01;R2day3-7=0.808,P<0.01).miR-21-5p levels were positively correlated with NIHSS scores(R2=0.728;P<0.01);positively correlated with 90-day mRS scores(R2=0.664;P=0.03).3.The bioinformatic analysis suggests that its target gene contained two biological process between the commissural neuron axon guidance(GO:0071679,p=0.077)and negative regulation of neuron apoptotic processes(GO:0043524,p=0.006).Conclusion:1.The expression levels of miR-21-5p in serum of patients with ICH were increased,which was significantly correlated with hematoma volume,perihematomal edema volume,the degree of neurological impairment and prognosis.2.miR-21-5p may be involved in the pathophysiological process of neurologic impairment and synaptic plasticity reconstruction after ICH. |
