| PartⅠ:The Correlation between Ghrelin and coronary artery calcificationObjective To observe the serum Ghrelin level in patients with coronary artery calcification and to analyze its correlation in coronary artery calcification.Methods Coronary atherosclerotic heart disease was diagnosed with concurrent coronary CTA,and 63 patients with calcified lesions in the coronary arteries were enrolled.61 patients with coronary atherosclerotic heart disease and coronary CTA diagnosis were not seen served as a control group.Compare the serum Ghrelin levels,serum calcium,Blood lipids(TG,TC,LDL-C),Analyze the correlation between serum ghrelin concentration and Ca2+,TG,TC,LDL-C and the risk factors of coronary artery calcification.Results The smoking,Ca2+,TG,TC,and LDL-C indexes of the two groups were statistically significant(P<0.05).There was no significant difference in age,gender,hypertension,diabetes,and body mass index(all P>0.05);The serum ghrelin concentration in patients with coronary artery calcification was lower than that in the control group(P<0.05);statistical analysis showed that serum ghrelin levels were negatively correlated with Ca2+,TG,TC and LDL-C.Age,smoking,Ca2+,TC,and LDL-C were risk factors for coronary artery calcification(P<0.05).Gender and TG had no significant effect on coronary artery calcification(all P>0.05).Ghrelin was coronary artery calcification.Protective factor(P<0.05).Conclusion Ghrelin is an independent protective factor for coronary artery calcification and can be used as an indicator to evaluate coronary artery calcification.PartⅡ:The role of Ghrelin in the vascular calcification by vitamin D3 combined with nicotine in vitroObjective To establish the rat vascular calcification model and explore the effect of Ghrelin on vascular calcification.Methods Forty sprague dawley rats aged 8 weeks were randomly divided into four groups:calcification group,control group,calcification+low concentration Ghrelin group,calcification+high concentration Ghrelin group.In the calcification group,nicotine was intragastrically administered(dissolved in peanut oil)and vitamin D3(dissolved in anhydrous ethanol)was intramuscularly injected,calcification+low concentration of Ghrelin group and calcification+high concentration of Ghrelin group was intraperitoneally injected with 50 ug/kg and 100 ug/kg Ghrelin once daily for 4weeks on the basis of vitamin D3 and nicotine treatment.The degree of calcification of rat thoracic aorta was observed by Alizarin red staining and Von Kossa section staining.Elisa method was used to measure serum calcium(Ca2+),alkaline phosphatase(ALP)and thoracic aortic bone morphogenetic protein-2,4(BMP-2,BMP-4)content,immunohistochemistry was used to detect the expression of BMP-2 and BMP-4 in the thoracic aorta.Western blot assay was used to detect the expression of four groups of BMP-2,BMP-4 and Ghrelin.Results The calcification group and the control group confirmed that the vascular calcification model was established successfully by Alizarin red staining and Von Kossa staining,and the high concentration of Ghrelin inhibited vascular calcification more than the low concentration of Ghrelin.The serum Ca2+,ALP,thoracic aorta BMP-2,BMP-4 content were measured by Elisa method.The BMP-2,BMP-4 protein expression confirmed that Ghrelin inhibited vascular calcification by immunohistochemistry and Western blot,high concentration of Ghrelin inhibited vascular calcification more significantly(P<0.05).Western blot was used to detect the expression of Ghrelin in the four groups,and there was no significant difference between the calcification group and the control group(P>0.05).Conclusion Nicotine combined with vitamin D3 can establish rat vascular calcification model,Ghrelin can inhibit the occurrence and development of calcification. |
