| Objective: The effect of autologous bone marrow mesenchymal stem cells transplantation on the repair of the anterior cruciate ligament injury in rabbits and the activity of urokinase type plasminogen activator,transforming growth factor β and white blood cells in rabbit serum were investigated.The expression level of interleukin-1 and interleukin-4Methods: 1.Sixty healthy adult male New Zealand white rabbits were selected and divided into normal group,model group and model bone marrow MSC transplantation group,with 20 rats in each group.2.The model group and the model bone marrow MSC transplantation group were used as the anterior cruciate ligament injury model.3.Acquisition and culture of autologous bone marrow MSC in rabbits and co-culture of bioprotein gel and bone marrow MSC.4.The model group and the model bone marrow MSC transplantation group were injected with a mixture of bio-protein glue,bone marrow MSC and bio-protein glue respectively;the normal group rabbits were not treated.5.The level of u-PA was detected by streptavidin-biotin-peroxidase complex(SABC)method.6.Detection of TGF-β,IL-1 and IL-4 levels by enzyme-linked immunosorbent assay.7.The WDW-100 microcomputer controlled electronic universal biomechanical testing machine was used to test the loading load when the anterior cruciate ligament was completely broken.Result:1.At the 2nd,4th,6th,and 8th week,the serum u-PA level gradually decreased,approaching the normal level,but the model bone marrow MSC transplantation group decreased significantly than the model group rabbit.The concentration of the rabbits in the model group was higher than that in the rabbits in the bone marrow MSC transplantation group,and the difference was statistically significant(P<0.05).2、At the 4th week,the serum TGF-β level of the rabbits in the model bone marrow MSC transplantation group was higher than that of the rabbits in the model group,and the difference was statistically significant(P<0.05).At the 2nd,6th and 8th week,the model group rabbits were higher than the model bone marrow MSC transplantation group and the normal group rabbit serum TGF-β levels,the difference was statistically significant(P<0.05).3、At the 2nd,4th,6th,and 8th week,the IL-1 level in the serum of the rabbits in the model group and the model bone marrow MSC transplantation group gradually decreased,approaching the normal level,but the model bone marrow MSC transplantation group decreased the amplitude of the rabbits larger than the model.Group of rabbits.The concentration level was higher in the model group than in the model bone marrow MSC transplantation group than in the normal group rabbits,the difference was statistically significant(P<0.05).4、At the 2nd,4th,6th,and 8th week,the IL-4 levels in the serum of the model group and the model bone marrow MSC transplantation group gradually decreased,which approached the normal level,but the model bone marrow MSC transplantation group decreased more than the model.Group of rabbits.The concentration level was higher in the model group than in the model bone marrow MSC transplantation group than in the normal group rabbits,the difference was statistically significant(P<0.05).5、At the 2nd,4th,6th,and 8th week,the injury site of the rabbits in the model group and the model bone marrow MSC transplantation group gradually improved,and the tensile force of the re-rupture gradually increased.The tensile force was larger thanthat of the normal group.Rabbits in the bone marrow MSC transplantation group were larger than the model group rabbits,and the difference was statistically significant(P<0.05).Conclusion: Autologous MSC transplantation after anterior cruciate ligament injury can reduce the concentration of u-PA,TGF-β,IL-1 and IL-4 in serum,an d promote the recovery of injury site. |
