Purpose:To observe the curative effect of the old Chinese medicine Chang Kang Fang on the rats with diarrhea-predominant irritable bowel syndrome.To study the regulation of Chang Kang Fang,on neurotransmitter in brain-intestinal axis of IBS-D model rats.To explore its influence on the brain-intestine interaction between CNS and ENS,and to inherit and dig the famous old Chinese medicine prescription.Methods:The model was constructed by combining senna leaf intragastric administration and binding stimulation as model method.After successful modeling,rats in the model group were randomly divided into five groups,model control group,high-dose group of Chang Kang Fang(H-CKF),medium-dose group of Chang Kang Fang(M-CKF),low-dose group of Chang Kang Fang(L-CKF),and positive drug group(pinaverium).Each group has ten rats,and the rats were given gavage for 14 days.To observe each group rats the improvement of IBS-D symptoms,and to take the colon tissue for pathological slices.The AWR score of each mouse after modeling and administration was detected,and the expression of brain-intestinal peptides ACTH,SP and VIP in the colon and brain tissues were detected by immunohistochemical method for semi-quantitative analysis.Results:(1)Comparing with the blank group,the AWR score of the modeling group was significantly increased(P<0.05)It suggested that the IBS-D rats was successfully established,combined with the general situation of rats,diarrhea,and intestinal mucosal pathology,and the model success rate was 76.92%.(2)After CKF and pinaverium treatment,compared w:ith the model control group,AWR scores of CKF group and positive group rats were decreased under various balloon pressures,showing statistical significance under balloon pressures of 10,20,40 and 60mmHg(P<0.05).It shows that CKF and positive drug group have showed some improvement in visceral hypersensitivity.(3)Compared with the blank group,the cells with positive expression of SP,VIP and ACTH in the brain tissue and intestinal tissue of the model control group were distributed in a relatively dense distribution,and the optical density OD value was increased.Compared with the model control group,the OD values of brain and intestinal peptides SP,VIP and ACTH in the CKF group and the positive group were all decreased,showing statistical significance(P<0.05),but there was no statistical significance between the concentrations.It shows that CKF group could reduce OD value of brain-intestinal peptide in IBS-D model rats.Conclusion:Chang Kang Fang can significantly improve the symptoms of diarrhea type irritable bowel syndrome model rats,reduce their intestinal hypersensitivity and the expression of brain intestinal peptide SP,VIP,ACTH.so it can be inferred that part of the mechanism of Chang Kang Fang used for the treatment of IBS-D may be affect the brain-intestine interaction between the central nervous system and the intestinal nervous system through the regulation of brain-intestinal neurotransmitters,then alleviate the symptoms of IBS such as abdominal pain and diarrhea. |
