Background and PurposeEsophageal cancer is one of the eight most common tumors in the world.More than 50% of the world’s esophageal cancer occurs in China,and its mortality and incidence rate ranks the fourth among Chinese malignant tumors.Malignant transformation of esophageal mucosa epithelium or glands leads to esophageal carcinoma.It is reported that about 400,000 people around the world suffer from esophageal cancer every year,and about half of them are Chinese.Similar to other gastrointestinal malignancies,esophageal malignant transformation is also a multi-stage,multi-step and multi-factor interaction process.In which,the activation of proto-oncogenes and the inactivation of tumor suppressor genes play an important role in the process of cell carcinogenesis.Therefore,a systematic study of the occurrence and development of esophageal cancer is of great significance for the early diagnosis of esophageal cancer,the improvement of the therapeutic effect of esophageal cancer and the guidance of the clinical prognosis of esophageal cancer.ZNF217 is a member of the zinc finger protein family,which is the largest transcription factor family found in the human genome.Different combinations and functional zinc finger sequences enable zinc finger proteins to perform multiple functions in biological processes,including development,differentiation,metabolism and autophagy.ZNF217 is a new candidate oncogene for epithelial neoplasm.Relevant studies have shown that ZNF217 is highly expressed in gastriccancer,ovarian cancer,prostatic carcinoma,colorectal cancer,breast cancer and so on.ZNF217 is closely related to the invasion and metastasis of tumor cells.ZNF217 is a member of the zinc finger protein family,and zinc finger protein is the largest transcription factor family found in the human genome.Different combinations and functional zinc finger sequences enable zinc finger protein to play a variety of functions in biological processes,including development,differentiation,metabolism and autophagy.Zinc finger protein 217(ZNF217)is a newly discovered candidate oncogene for epithelial tumors.Relevant studies have shown that ZNF217 is highly expressed in gastric cancer,ovarian cancer,anterior adenocarcinoma,colorectal cancer,breast cancer and other tumor tissues,and is closely related to the invasion and metastasis ability of tumor cells.However,after reviewing a large number of literature,there have been no relevant reports on the expression of ZNF217 in esophageal cancer tissues,its expression level and its practical clinical significance.In this study,q-pcr and immunohistochemistry(IHC)were used to explore the difference between esophageal cancer and paracancer tissue,and statistical analysis was performed to study its clinical significance.Methods1.Collection of clinical specimens:(1)Fresh specimens were collected from 50 patients with esophageal squamous cell carcinoma in linzhou tumor hospital on May1,2018-August 31,2018.Fresh specimens were collected after thoracic surgery in linzhou tumor hospital,esophageal cancer tissue and adjacent normal tissue were cut and placed in cryopreservation tube and stored in liquid nitrogen tank for later use.(2)2018.09.01-2018.09.30 consulting cases of patients with esophageal squamous cell carcinoma in the medical record department of the first affiliated hospital of zhengzhou university,this case was a total of 998 patients who visited the first affiliated hospital of zhengzhou university on January 1,2012 and underwent esophageal squamous cell carcinoma surgery on May 31,2013.The patient information was registered.At last,89 cases were successfully followed up,the wax blocks of esophageal squamous cell carcinoma and para-carcinoma tissues wereborrowed from the department of pathology of the first affiliated hospital of zhengzhou university and sliced for later use.2.mRNA was extracted from 50 fresh frozen esophageal squamous cell carcinoma tissues and the corresponding para-carcinoma tissues,and the mRNA was reversely transcribed into cDNA.The expression level was detected by real-time fluorescence quantitative PCR(qRT-PCR).3.The expression level of ZNF217 protein in esophageal squamous cell carcinoma and adjacent normal tissues was detected by immunohistochemistry(IHC).4.SPSS21.0 software was used for statistical analysis of experimental data.Paired T test was used to analyze the mRNA and protein expression levels of esophageal cancer tissues and adjacent tissues,and chi-square test was used to analyze the relationship between mRNA and protein expression levels with clinicopathology.Kaplan-meier method was used to plot the survival curve.Results1.The expression level of ZNF217 mRNA in the esophageal squamous cell carcinoma was higher than that in the adjacent tissues,and the results showed statistical difference(P=0.0018).The expression level was related to lymph node metastasis(P=0.018)and tumor stage(P=0.023).2.The expression level of ZNF217 protein in esophageal squamous cell carcinoma was higher than that in the adjacent tissues,and the results showed statistical difference(P=0.0018).The expression level was related to tumor size(P=0.015),lymph node metastasis(P=0.000),invasion depth(P=0.002)and tumor stage(P=0.008).3.The 5-year survival rate curve showed that the 5-year survival rate of patients with esophageal squamous cell carcinoma with positive ZNF217 expression was significantly lower than that with negative ZNF217 expression(P=0.0137).ConclusionsZNF217 mRNA and protein expression were up-regulated in esophageal squamous cell carcinoma,and were related to the clinical and pathological characteristics of patients with malignant tumor.ZNF217 may be an oncogene of esophageal squamous cell carcinoma,which can promote the growth and migration of tumor cells and play a positive regulatory role in the occurrence and development of esophageal carcinoma.ZNF217 may have clinical and scientific value in esophageal cancer,which can help esophageal cancer with early diagnosis and molecular targeted therapy. |
