Objective:To analyze the relationship between the expression of programmed cell death 4(programmed cell death 4,PDCD4)in four human breast cancer cell lines MCF-7、T47D、SKBR-3、MDA-MB-231 and their migration ability and invasion ability in vitro,the effect of PDCD4 gene on the development of breast cancer was preliminarily explored.Methods:1.The expression of PDCD4 gene in normal human mammary epithelial cell line MCF-10 A and four kinds of human breast cancer cell lines MCF-7、T47D、SKBR-3 and MDA-MB-231 were detected by quantitative real-time PCR(q RT-PCR).2.Western-blot was used to detect the expression of PDCD4 gene in normal human mammary epithelial cell line MCF-10 A and four kinds of common breast cancer cell lines MCF-7、T47D、SKBR-3 and MDA-MB-231 from the protein level.Combined with the experimental results of q RT-PCR,the relationship between them was analyzed,thereby screening out the two breast cancer cell lines with low PDCD4 expression and high expression.3.The invasion ability of four kinds of breast cancer cell lines of MCF-7、T47D 、 SKBR-3 and MDA-MB-231 were detected by Transwell chamber experiment,and the relationship between PDCD4 gene and its invasion ability was analyzed.4.The migration ability of four kinds of breast cancer cell lines of MCF-7、T47D、SKBR-3 and MDA-MB-231 were detected by scratch experiment,and the relationship between PDCD4 gene and its migration ability was analyzed.Results:1.The expression of PDCD4 m RNA was highest in normal human mammary epithelial cell line MCF-10A(1.000±0.000),in MCF-7(0.825±0.103)、T47D(0.674±0.086)、SKBR-3(0.438±0.071)and MDA-MB-231(0.270±0.038),the expression levels of PDCD4 m RNA in the four breast cancer cell lines were sequentially decreased,and the expression levels of PDCD4 m RNA in these five cell groups were compared,the difference is statistically significant(p<0.05).2.The expression of PDCD4 protein was highest in normal human mammary epithelial cell line MCF-10A(1.094±0.047),in MCF-7(0.863±0.072)、T47D(0.627±0.105)、SKBR-3(0.450±0.044)and MDA-MB-231(0.335±0.058),the expression levels of PDCD4 protein in these four breast cancer cell lines were sequentially decreased,and the expression levels of PDCD4 protein in these five cell groups were compared,the difference is statistically significant(p<0.05).3.The expression of PDCD4 m RNA and PDCD4 protein in these five cell groups was positively correlated(r=0.963,P<0.001).4.The number of the four kinds of breast cancer cell lines measured by Transwell chamber were: MCF-7(20±8),T47D(43±8),SKBR-3(68±12)and MDA-MB-231(97±13);The scratch healing rates of the four breast cancer cell lines detected by the scratch test were: MCF-7(16.67±4.22),T47D(25.00±3.00),SKBR-3(33.33±4.25)and MDA-MB-231(46.15±3.44)Comparing the invasion and migration abilities between the four breast cancer cell groups,the differences were statistically significant(P<0.05).And the migration ability and invasion ability of these four kinds of breast cancer cells were positively correlated(r=0.918,P<0.001).5.The expression of PDCD4 m RNA and PDCD4 protein in these four kinds of human breast cancer cells was negatively correlated with their invasion ability,and the correlation coefficients were(r=-0.769,p=0.004<0.05;r=-0.729,p=0.007<0.05).Moreover,the expression of PDCD4 m RNA and PDCD4 protein in these four kinds of human breast cancer cells also showed a negative correlation with their migration ability in vitro,and the correlation coefficients were(r=-0.861,P<0.001;r=-0.856,P<0.001).Conclusions:1.The expression of PDCD4 gene in normal human mammary epithelial cell lines and four common breast cancer cells was: MCF-10A>MCF-7>T47D>SKBR-3> MDA-MB-231.2.The migration ability and invasion ability in vitro of the four common human breast cancer cells were ranked as follows:MCF-7<T47D<SKBR-3<MDA-MB-231.There was an obvious positive correlation between migration ability and invasion ability,both of which could represent the proliferation and metastasis ability of malignant tumor cells.3.The expression of PDCD4 gene in breast cancer cells had a significant negative correlation with its invasion and migration ability.It was suggested that PDCD4 gene might be involved in the biological behavior of breast cancer cell proliferation and metastasis,which provided clues to the molecular mechanism of breast cancer development.
