| Objective: To investigate the relationship between PAX-8,WT-1,Ki-67,P53,Vimentin,ER and PR in immunohistochemical indexes of epithelial ovarian cancer and the clinicopathological characteristics,drug resistance,metastasis and prognosis of ovarian cancer patients.To Provide a basis for personalized treatment and prognosis of ovarian cancer patients.Methods: We retrospectively reviewed 191 patients with epithelial ovarian cancer who were treated with complete the initial treatment in the Affiliated Tumor Hospital of Guangxi Medical University from January 2011 to December2016.According to our department in the postoperative pathological immunohistochemical report issued by the PAX-8,Ki-67,WT-1,P53,Vimentin,ER and PR as a result,Kaplan-Meier was used to describe the survival curve,and Log-rank test,χ2 test independent sample T test,one-way ANOVA for univariate analysis,logistic regression model and Cox regression model were used for multi-factor analysis,and decision tree method was used to screen immunohistochemical index combinations.Results: 1.Single factor analysis of each immunohistochemical index and clinicopathological features showed that the index related to the age of the patient was the ratio of P53 positive cells;the index associated with FIGO stagewas WT-1 positive expression and Vimentin negative expression;The relevant pathological types of ovarian cancer are PAX-8,WT-1,Vimentin,ER and PR expression;the indicators associated with distant metastasis of ovarian cancer are WT-1 positive expression and PR negative expression;and the relevant indicators associated with ovarian cancer tissue differentiation were PAX-8positive expression,WT-1 positive expression,Vimentin negative expression and PR positive expression,WT-1 low expression and P53 positive expression cell ratio;positive expression of WT-1 and low expression of PR were correlated with lymph node metastasis.2.The immunohistochemical indexes and patient’s OS and PFS single factor analysis showed that the positive expression of WT-1 was significantly correlated with PFS of patients.There was no significant difference in the expression of other immunohistochemical indexes and OS and PFS.3.Cox multivariate regression results showed that no immunohistochemical indicators were included in the prognosis and recurrence model.After classification of immunohistochemical markers,logistic regression analysis showed no significant results in patients with recurrence,metastasis,prognosis and drug resistance.4.Decision tree classification analysis results: 1)The combination of immunohistochemical markers associated with recurrence was low expression of Ki-67 and positive PR expression.The PFS of patients who shared this combination in cancer tissues was longer than that of other patients in this group,and the difference was statistically significant.2)The combination of metastasis-associated immunohistochemical markers was: WT-1 positive,ER positive,and PR negative.The proportion of distant metastasis in patients who shared this combination in cancer tissues was lower than that in other patients inthe group.The difference was statistically significant.3)The prognosis-related combination was WT-1 positive,ER positive and Ki-67 high expression and WT-1 positive and ER positive expression.Patients with WT-1 positive,ER-positive,and high Ki-67 expression in cancer tissues,OS within 2 years shorter than other patients in this group,WT-1 positive and ER-positive patients co-expressed in cancer tissues,OS within 3 years shorter than other patients in this group,the difference was statistically significant.Conclusion: 1.PAX-8 expression is positively correlated with poor differentiation of ovarian cancer tissues and is associated with pathological types of ovarian cancer.2.WT-1 expression was associated with pathological type of ovarian cancer,FIGO stage,and its positive expression was associated with low degree of cancer tissue differentiation,distant metastasis,and lymph node metastasis.Low expression of WT-1 is associated with high differentiation in cancerous tissues.Patients with positive expression of WT-1 had shorter progression-free survival and ovarian cancer was more likely to relapse.3.Vimentin expression was associated with FIGO stage and pathological type of ovarian cancer,and Vimentin negative expression was positively correlated with poor differentiation of ovarian cancer tissues.4.ER and PR had the highest positive expression rate in endometrioid carcinoma,and ER had the highest expression rate in endometrioid carcinoma.The positive expression of PR was positively correlated with the high differentiation of ovarian cancer tissues.The positive expression of PR was negatively correlated with distantmetastasis,and the low expression of PR was positively correlated with lymph node metastasis.5.The positive expression rate of P53 was positively correlated with the age of disease and the poor differentiation of ovarian cancer tissues.6.Patients with low expression of Ki-67 and positive PR expression in cancer tissues had longer progression-free survival and a better prognosis.Patients with WT-1 positive,ER positive,and PR negative in cancer tissues are less likely to develop distant metastasis of ovarian cancer.Patients with WT-1 positive,ER-positive,and high Ki-67 expression in cancer tissues,and patients with WT-1 positive and ER-positive in cancer tissues,had shorter overall survival and worse prognosis. |
