| Objective:The expression and difference of autophagy-related genes LC3,Raptor,Rictor and Beclinl in gastric mucosa,chronic atrophic gastritis,low grade intraepithelial neoplasia,high grade intraepithelial neoplasia and early gastric cancer were detected by immunohistochemistry.The relationship between LC3,Raptor,Rictor and Beclinl in the development and progression of early gastric cancer was analyzed.The relationship between LC3,Raptor,Rictor and Beclinl in the development and progression of early gastric cancer was analyzed.The results provided a new idea for the diagnosis and treatment of early gastric cancer.Materials and MethodsA total of 160 ESD specimens from endometriosis affiliated hospitals of Binzhou Medical College from June 2015 to December 2016 were selected,including 30 cases of normal gastric mucosa,chronic atrophic gastritis,low grade intraepithelial neoplasia and high grade intraepithelial neoplasia,40 cases of early gastric cancer;85 males and 75 females;aged 30-70 years old,the median age of 50 years;According to the consensus of early gastric cancer screening and endoscopic diagnosis and treatment in China(2014,Changsha)(main reference to Vienna classification criteria for intraepithelial neoplasia),the selected cases were double-blinded and re-examined by two experienced pathologists.All cases were found for the first time,without any preoperative radiotherapy and chemotherapy.Immunohistochemistry EnVision staining,LC3,Beclinl with positive sections of known colorectal cancer as a positive control,Raptor,Rictor with testicular sections as a positive control,PBS instead of primary antibody as a negative control.LC3 was expressed in the cell membrane and cytoplasm;Raptor,Rictor and Beclinl were expressed in the cytoplasm,the positive signs of brown particles appear,each of the 10 randomly selected high magnification(400×)field of vision image acquisition using Image Pro Plus 6.0 image analysis software to detect the integrated optical density(IOD)values of the 10 positive regions of the selected field.The mean values of the expression values of LC3,Raptor,Rictor and Beclinl.SPSS 23.0 software was used for statistical analysis.Themeasurement data were expressed as mean±standard deviation(x±s).One-way analysis of variance(ANOVA)or independent sample t-test was used to compare the two groups.Spearman correlation analysis was used to analyze the correlation between protein expression,Correlation with clinical and pathological factors using multiple linear regression analysis.ResultsThe expression of LC3 in each group gradually increased with the progression of lesion degree,except for the low grade intraepithelial neoplasia and atrophic gastritis group,there was no significant difference between the expression of LC3 and non-tumorous gland Membrane,while in the tumor glands are expressed in the membrane and cytoplasm;The expressions of Raptor and Rictor in early gastric cancer,high grade intraepithelial neoplasia and low grade intraepithelial neoplasia were higher than those in atrophic gastritis and normal mucosa,but the expression of Raptor in early gastric cancer was lower than that in intraepithelial neoplasia.The expressions of Raptor and Rictor in early gastric cancer,high grade intraepithelial neoplasia and low grade intraepithelial neoplasia were higher than those in atrophic gastritis and normal mucosa,but the expression of Raptor in early gastric cancer was lower than that in intraepithelial neoplasia;While the expression of Rictor increased gradually with the increase of tumor grade;the expression of Beclinl in normal mucosa and each tumor group was lower than that in atrophic gastritis group,and its expression gradually decreased with the increase of tumor grade.In early cancer group,almost not express.Conclusion1.LC3,Raptor,Rictor and Beclinl expression in normal mucosa,chronic atrophic gastritis,low-grade intraepithelial neoplasia and high-grade intraepithelial neoplasia and early gastric cancer group,play an important role in the occurrence and development of early gastric cancer Character.2.The expression levels of autophagy-related factor LC3,Raptor and Rictor differed in the occurrence and development of early gastric cancer,and the coordination of the three involved in the occurrence and development of early gastric cancer.3.The expression of autophagy-related gene LC3 in the non-tumorous gland mainly in the cell membrane,but in the tumor gland expression in the cell membrane and cytoplasm,therefore,speculated that LC3 can be used as a secondary diagnosis of early gastric cancer marker things.4.The correlation between autophagy-related factor LC3 and Raptor and Rictor was related to the differentiation and invasion depth of early gastric cancer,suggesting that the three factors are closely related to the development of early gastric cancer... |
