The Metabonomic Research About The Effect Of Different Boiling Time Of Xiaoqinglong Decoction In The Treatment Of Bronchial Asthma

Objective: the first part,confirm the boiling time of Xiaoqinglong Decoction as the discription in Shang Han Lun.The second part,determine the influence on Xiaoqinglong Decoction constituted of 9g asarum of different boiling time based on LC/MS technique to compare the difference between different time,and explore the mechanism treatment of asthma with Xiaoqinglong Decoction.Methods: the first part: converting the dosage and water yield of Xiaoqinglong Decoction according literature,and boil three times as discription in Shang Han Lun,and calculate average to determine the boling time.the second part: 48 SD rats were randomly divided into normal group,model group,positive control group,different boling time of Xiaoqinglong Decoction including 15 minutes,25minutes,35 minutes,45 minutes and 55 minutes.Establish the asthma model in two steps except the normal group with sensitization and motivation.And the normal group and model group were perfused with saline,the positive group was perfused with aminophylline,other groups were perfused with Xiaoqinglong Decoction of different boling times.After that,collecting blood in abdominal aorta and hold the serum to check by LC/MS technique.Result: 1.the decocting time of Xiao Qinglong soup was about 45 minutes according the original decoction method,while the modern time is 35 minutes.2.The PCA results of Xiaoqinglong Decoction in 35 minutes group were closer to the normal group compared with other groups based on LC/MS technique,and the effect was the best.3.We identified 14 kinds of potential markers which involved four highways including Glycosylphatidylinositol-anchor biosynthesis、Sphingolipid metabolism、Lysophosphlipid metabolism and Glycerophospholipid metabolism associated with Xiaoqinglong Decoction in the treatment of asthma in rat serum,the results showed that Xiaoqinglong Decoction has callback function on the 14 kinds of markers,So Xiaoqinglong Decoction is to regulate lipid metabolize by callback the content of potential markers to cure inflammation,bronchoconstriction and airway remodeling.Conclusion: 1.The original decoction time of Xiaoqinglong soup is about 45 minutes through physical measurement.2.The 45 minute Xiaoqinglong Decoction was the best treatment compared with the other groups based on the successful asthma model.3.Xiaoqinglong Decoction is to regulate lipid metabolize by callback the content of potential markers to cure inflammation,bronchoconstriction and airway remodeling by analysing the potential markers and highways based on LC/MS technique.And it illustrate that the treatment mechanism is probably correlative to lipid metabolize.
