Objective:Prostate cancer PCa is a common male disease,in Europe and the United States,PCa incidence is very high,the health of men who have serious harm.Early diagnosis of the disease is very important,early PCa by taking radical means,the 5-year survival rate can reach 100%,while for advanced and multiple metastases in patients,the 5-year survival rate of only 32%.For a long time,clinically,mainly through PSA screening and transrectal B-guided biopsy of PCa for diagnosis,but the limitations of the larger,the diagnostic effect is not ideal.The modern medical study found that more than 80% of PCa patients with the body androgen regulatory gene transmembrane serine protease(Transmembrane Prostease Serine 2 TMPRSS2)and ETS family of genes(such as ERG,ETV1,ETV4)fusion,and this The phenomenon is not seen in benign prostate disease.Fluorescence In-Situ Hybridization(FISH)is a detection technique based on basic complementary pairing.The known single-stranded nucleic acid is used as a probe to detect specific DNA sequences.FISH has the advantages of simple operation,high sensitivity,accurate detection and so on.This study explored the significance of TMPRSS2-EG fusion gene in the diagnosis and diagnosis of prostate cancer by detecting the fusion of genes in TMPRSS2 and ETS family(such as ERG,ETV1,ETV4),analyzing the correlation with the clinical diagnostic parameters of PCa.Methods : In this project,30 cases of postoperative pathological samples of prostate were selected,including 15 samples of radical prostatectomy and 15 cases of benign prostatic hyperplasia.Patients were not treated with radiotherapy and chemotherapy before surgery.All specimens were fixed with 10% formalin and embedded in paraffin.The routine pathological examination was performed.FISH was used to detect the fusion of TMPRSS2 and ETS family gene ERG.The clinical data were analyzed and the results were analyzed.(TRUS)and rectal examination(DRE),isotope full-body scan(ECT)were used to compare the relationship between TMPRSS2-ETS fusion gene and GMPason score.Results:Through the detection of pathological specimens of patients,the results are as follows:(1)30 cases of pathology specimens,15 cases of TMPRSS2-ETS family gene fusion,and all of the ERG fusion.15 cases of positive specimens of pathological diagnosis,diagnosis of 15 cases of all prostate cancer.(2)15 cases of TMPRSS2-ERG fusion gene were negative,the TMPRSS2-ETV1 and TMPRSS2-ETV4 fusion gene showed negative.Of the 15 cases of negative patients with pathological diagnosis,all were diagnosed with benign prostatic hyperplasia.(3)The sensitivity of prostate cancer was 89.3%(25/28),the specificity was100%,the positive predictive value was 100% and the negative predictive value was84.2%(16/19)by using TMPRSS2-ERG fusion gene.(4)The abnormal state of TMPRSS2-ETS fusion gene was positively correlated with Gleason score and ECT(P=0.001,r=0.383;P=0.041,r=0.306).The incidence of TMPRSS2-ETC fusion gene was not correlated with the positive rate of DRE and TRUS(P> 0.05).Conclusion:TMPRSS2-ETS fusion gene mutation was detected by FISH,and the specificity and sensitivity of the TMPRSS2-ETS gene were positively correlated with the Gleason score and ECT outcome of the patients.Therefore,the detection of TMPRSS2-ETS gene fusion diagnosis of PCa can improve the accuracy of clinical diagnosis,for PCa patients with radical treatment is an important help. |
