| BackgroundAs one of the main complications of diabetes,Diabetic Nephropathy(DN)is a common cause of chronic renal failure in the world.The prevalence of the disease is increasing.Now,DN is no longer simply considered as a metabolic disease,but a progressive chronic inflammatory disease,but the specific mechanism of the activation of inflammatory factors is still unclear,among which the study of epigenetics has become a new focus.As an important anti-inflammatory medium,IL-10 has both anti-inflammatory and immunosuppressive effects,and its activity is closely related to the clinical prognosis of DN.ObjectiveTo detect the expression of Interleukin IL-10 and DNA methylation transferase-1(Dnmt-1)in renal tissue of patients with Type 2 Diabetic Nephropathy(DN),then investigate the relationship between the two indicators and analyze their relationship with various clinical indicators.Methods23 cases of renal biopsy diagnosed as DN were chosen as experimental group,and 9 cases pathologically diagnosed as Mild Lesion Glomerular Disease were chosen as control group.The expression of IL-10 Dnmt-1 was monitored by Immunohisto-chemistry,the difference in the expression was compared and the relationship between the two indicators and clinical data in DN patients was analyzed.Result1.Compared with the control group,DN group showed significant obvious characteristics of proteinuria,higher cystatin C and blood lipids,faster erythrocyte sedimentation,lower serum albumin,and red blood cell counts.2.IL-10 was expressed in both DN group and control group,and mainly expressed in renal tubular cytoplasm.The Mann-Whitney U test of Non-parametric showed: z=-0.315,p=0.752,(P>0.05).There was no significant difference between DN group and control group.3.The expression of Dnmt-1 in DN group was significantly higher than that in control group.In DN group,the expression was mainly in nucleur of renal tubular cells.Median(interquartile spacing)results showed that Dnmt-1 positive cell count of the former was 43.0(31.0-48.0),while the latter was 0.3(0.05-0.35).The Mann-Whitney U test showed z=-4.434,p=0.000(P<0.05).The difference was statistically significant.3.There was significant negatively correlation between the expression of IL-10 and Dnmt-1 in DN group(r=-0.434,p=0.038)(P<0.05).The difference was statistically significant.ConclusionThe expression of the anti-inflammatory factor IL-10 could be detected in the renal tissue of Type 2 Diabetic Nephropathy,and the secretion of IL-10 was negatively correlated with the expression of Dnmt-1. |
