Study On The Effect Of "Tiao Shen Chang Qing" Acupuncture Method On NP-Y、Ghrelin In Serum And Brain Tissue Of PDD Model Rats

Objective: The effects of Tiao Shen Chang Qing acupuncture and routine acupuncture on the content of brain and intestinal peptide NP-Y,Ghrelin in serum and brain tissue of rats with Parkinson’s disease with depression were compared,and the regulatory effects of different intervention methods on NP-Y,Ghrelin were discussed.Methods: The rat model of Parkinson’s disease(PD)was established by 6-OHDA unilateral striatum double target injection.The success rate of PD was verified by APO induced behavioral test.After successful modeling,the rat model of PDD was established on the basis of PD model combined with CUMS.Weight measurement,open field test and 1% sucrose solution preference test were used to detect whether the model of PDD was successful.The rats were randomly assigned to PDD model group,drug control group,Tiao Shen Chang Qing acupuncture group,routine acupuncture group,false operation group and blank group with 10 rats in each group.The drug group was treated with Madopar and fluoxetine intragastrically.The Tiao Shen Chang Qing acupuncture group was treated with Tiao Shen Chang Qing method acupuncture,the routine acupuncture group was treated with modern acupuncture at high frequency acupoints of PDD,and the blank group was raised normally and not treated.Behavioral tests were performed 28 days after treatment.After behavioral examination,the serum and brain tissue of rats in each group were taken,and the expression of NP-Y,Ghrelin in serum and brain tissue was detected by ELISA method.Statistical analysis of the data with SPSS17.0 software.Results: Compared with the blank group,there was no significant change in the content of NP-Y,Ghrelin in serum and brain tissue in the pseudo-operation group(P > 0.05),The behavioral indexes of the model group were significantly different(P < 0.01),and the content of NPY,Ghrelin was significantly decreased(P <0.01).Compared with the model group,the behavioral indexes of the drug control group were significantly different(P < 0.01),but the content of NPY,Ghrelin had no significant change(P >0.05).Compared with the model group,the behavioral indexes and the content of NPY,Ghrelin in the routine acupuncture group and Tiao Shen Chang Qing acupuncture group were significantly higher than those in the model group(P<0.05).Compared with the drug control group,the behavioral indexes of routine acupuncture group were statistically significant(P<0.05),but there was no significant difference between drug group and Tiao Shen Chang Qing group(P>0.05);Compared with the drug control group,the changes of NPY,Ghrelin content in the routine acupuncture group and Tiao Shen Chang Qing acupuncture group were significantly different(P < 0.05).Compared with the routine acupuncture group,there was significant difference in behavioral indexes between the routine acupuncture group and Tiao Shen Chang Qing acupuncture group(P < 0.05),the content of NPY,Ghrelin was significantly increased(P < 0.05).Conclusion:1.In improving the behavior of PDD model rats,the acupuncture method of "Tiao Shen Chang Qing" can basically achieve the effect of drugs,and it is superior to conventional acupuncture.2.The contents of NP-Y,Ghrelin in serum and brain tissue of PDD model rats were significantly increased by "Tiao Shen Chang Qing" acupuncture and routine acupuncture,and the acupuncture method of "Tiao Shen Chang Qing" was better than that of conventional acupuncture.The effect of drugs on the above indexes is not obvious.3.One of the mechanisms of the "Tiao Shen Chang Qing" method may be to regulate the content of NP-Y and Ghrelin in the serum and brain tissue to play a role.
