Objective: This study was to investigate the inhibitory effect of pirfenidone on rat incision scar formation and its possible mechanism of action,and provide a new direction for the research and development of new treatment methods for scar.Materials and Methods: Thirty-two male SD rats of normal grade were selected and healthy,weighing about 200 g,provided by the Experimental Animal Center of Anhui Province.Rats were housed in a single cage during the experiment.Thirty-two male Sprague-Dawley rats were anesthetized with 10% chloral hydrate(by laboratory configuration)before surgery.After anesthesia,the back hair of the rats was shaved.After shaving,two longitudinal parallel incisions were made on both sides of the spine.It is about 1.5 cm long and is deep into the fascia.The incision is not sutured and allowed to heal itself.The rats were kept in a single cage and 1 cage was used to avoid biting the incisions between the rats.4 incisions per rat,each incision can be considered as a separate sample A total of 128 incisions were made in 32 rats and divided into 4 groups: blank control group(no treatment during the experiment);negative control group(external blank cream matrix);4% pirfenidone cream Group;8% pirfenidone cream group.Four nicks on each side of each rat were divided into different groups.The dressing was changed once a day,about 0.5 g at a time,and the rats were normally bred,and the rats were anesthetized 14 days later.Scar tissue specimens were fixed,fixed in 10% formalin,embedded,sectioned,and subjected to HE staining and Masson staining,respectively.Results: 1.According to the results of Elisa assay,the expression level of TGF-β1 in the experimental group was significantly lower than that in the control group,and the lower concentration of TGF-β1 in the high concentration experimental group was further lower.2.According to the HE staining count,the number of fibroblasts in the experimental group was significantly lower than that in the control group,and the concentration of fibrocytes in the high concentration experimental group was further reduced in the experimental group.3.According to the Masson staining microscope,the color of the collagen fiber staining in the control group was obvious,the fiber arrangement was disordered and coarse,while the collagen fibers in the experimental group were arranged neatly and the shape was slender.Conclusions: This study showed that the post-traumatic incision scar tissue immediately applied pirfenidone cream to reduce the expression of TGF-β1 in the scar formation process of the experimental group,thereby inhibiting the proliferation of fibroblasts,inhibiting collagen synthesis,and finally inhibiting.The effect of scar formation,but will extend the healing time of the incision.
