| Objective:Cholangiocarcinoma(CCA)belongs to a type of primary liver cancer.Due to the complex phenotypes and different cell sources of CCA,it has tissue diversity.Currently,CCA is diagnosed mainly by Examination and puncture pathologieal biopsy.However,due to its occult occultity and difficulty in diagnosis,CCA may have missed the best treatment opportunity once it is discovered,and the prognosis is poor.The study found that the incidence of CCA has been increasing year by year since the 21st century,and it has seriously threatened human health.People are paying more and more attention to the research on the pathogenesis of CCA.The process of carcinogenesis is complicated and involves the disorder of specific cell genomes.Therefore,exploring the biological characteristics of CCA from the gene level has become one of the hotspots of current research.The tumor suppressor gene TIP30 is a newly discovered tumor suppressor gene that has the ability to promote apoptosis,inhibit tumor angiogenesis,inhibit tumor cell proliferation,and inhibit cell invasion.It is used in tumors such as breast cancer,lung cancer,liver cancer,and pancreatic cancer.It plays an important role in the occurrence and development In CCA,relevant studies and previous experiments in our research group showed that TIP30 was down-regulated in CCA tissue samples and correlated with clinical stage,metastasis and survival prognosis(P<0.05).However,the specific molecular mechanism by which TIP30 plays a role in CCA has not been reported yet.The purpose of this study was to explore the effect of TIP30 on the expression of related signaling pathways in cholangiocarcinoma.Method:1.Using RNA interference technology to silence the expression of TIP30 in cholangiocarcinoma cell lines RBE1,HUCCT1,9810cells and verify the transfection efficiency by qRT-PCR and Western blot.2.Using qRT-PCR to detect changes in NGF/TRKA signaling pathway and BCL-2 gene in RBE1,HUCCT1,and 9810 cells after interference with TIP30 expression.3.Using qRT-PCR and Western Blot to detect the changes of NOTCH signaling pathway,WNT/β-catenin signaling pathway,in RBE1,HUCCT1,9810 cells after interference with TIP30 expression.Results:1.Down-regulation of TIP30 does not affect the changes of NGF/TRKA pathway,NOTCH signaling pathway,WNT/β-catenmn signaling pathway in RBE1,HUCCT1 and 9810 cells.2.Down-regulation of TIP30 can promote the mRNA expression of BCL-2 gene in RBE1,HUCCT1 and 9810 cells.Conclusions:Down-regulation of TIP30 can promote the mRNA expression of BCL-2 gene in cholangiocarcinoma... |
