Background:Stroke is a cerebrovascular disease with a very high mortality rate and disability.Ischemic stroke accounts for about 80% of strokes,and there are currently no specific treatments.With the development of high-throughput sequencing technology,it has been found that long-chain non-coding RNA(LncRNA)that has been neglected can be made into a process of ischemic stroke through various molecular biology machines,but LncRNA and ischemic The specific relationship of stroke is currently reported less.In this study,we explored the differential expression profiles of LncRNA in the serum of patients with acute ischemic stroke,and conducted bioinformatics analysis to explore possible mechanisms.This study enriches the biological function of LncRNA and provides a potential diagnostic and therapeutic target for ischemic stroke at the genetic level.Methods: Using high-throughput sequencing technology,20 patients with acute ischemic stroke who met the inclusion criteria were sequenced,and LncRNA with significant difference in expression level was searched based on FPKM values.Then,some differentially distinct LncRNAs were further verified by qPCR technique,and according to Its proximity in spatial position,searching for LncRNA related genes,performing GO analysis and KEGG pathway analysis,predicting the signal pathway and function of LncRNA with significant differences,and searching for abnormally expressed LncRNA in patients with acute ischemic stroke The role of.Result: A total of 44578 LncRNAs were detected,of which 228 were up-regulated and16 were down-regulated.GO analysis showed that 184 entries were obtained in patients with ischemic stroke.Among them,the up-regulated expression of LncRNA involves 173 items of biological pathway(BP),1 item of cell component(CC),and 7 items of molecular function(MF).The down-regulated expression of LncRNA involved 3 entries in BP,with no CC and MF terms.KEGG PATHWAY pathway analysis showed that the down-regulatedexpression of LncRNA did not have an enriched signaling pathway,while the up-regulated group had Glutathione metabolism,PPAR signaling pathway,Type II diabetes mellitus,Aminoacyl-tRNA biosynthesis,Drug metabolism-cytochrome P450,Adipocytokine signaling pathway,Metabolism of xenobiotics by cytochrome P450,Chemical carcinogenesis,Longevity regulating pathway-mammal,AMPK signaling pathway,Axon guidance,Cell adhesion molecules(CAMs),Non-alcoholic fatty liver disease(NAFLD),Viral carcinogenesis,HTLV-I infection.The expression of 6 groups of LncRNA(ENSG00000269900,ENSG00000196559,ENSG00000202198,ENSG00000226482 and ENSG00000260539 for up-regulation and XLOC0139942 for down-regulation)was confirmed by qPCR.It has significant significance and research value.Conclusion: High-throughput sequencing technology has found that a large number of LncRNAs that are up-regulated or down-regulated in patients with acute ischemic stroke have been shown to have the same directionality as high-throughput sequencing results by validating partial LncRNA.GO analysis and KEGG PATHWAY pathway analysis showed that there are many items and pathways that have significant significance and research value.This study found that in the pathological process of patients with acute ischemic stroke,the up-regulation of ENSG00000226482 may affect the progression of the disease by activating the adipocytokine signaling pathway,providing new treatment and idea for acute ischemic stroke and prognosis.
