Purpose : To study on the effects of dexmedetomidine on sevoflurane-induced apoptosis in rat hippocampal neurons on the basis of inhaled sevoflurane,and to provide more theoretical basis for reducing the damage of nerve cells in clinical anesthesia.Methods:Healthy male SD rats were randomly divided into four groups: sham operation group(group A),sevoflurane group(group S),dexmedetomidine +sevoflurane group(D+S group),yohimbine + right Metopidine + sevoflurane group(Y+D+S group).Each group was labeled,Y+D+S group was intraperitoneally injected with yohimbine 2.5 mg/kg,dexmedetomidine 20 ug/kg,and 1.5% sevoflurane was inhaled for 1 h/d.D+S group dexmedetomidine 20 ug/kg,inhaled 1.5% sevoflurane 1h/d.Group S inhaled 1.5% sevoflurane for 1 h/d.Group A animal experiment center was raised at room temperature.After 14 days,Y-maze was used to detect the learning and memory ability of rats in each group.HE staining technique and Nissl staining technique were used to observe the morphological changes of rat hippocampal neurons.The expression levels of Caspase-3,Bax and Bcl-2 in hippocampus of rats were determined by immunohistochemistry and Western blot.Result:Y-maze test showed that the spatial learning and memory ability of SD rats in S group and Y+D+S group decreased significantly.The spatial learning and memory ability of SD rats in D+S group also decreased,but the degree of decline was smaller than that of S group and Y+D+S group.Immunohistochemistry and Western blot analysis showed that the expression of Caspase-3 and Bax in group A and D+S was less than that in group S and Y+D+S,while that of Bcl-2 was higher than that in group S and Y+D+S.many.In the D+S group,HE staining of hippocampal neurons showed less cell shrinkage and more complete morphology.The hippocampal nerve cells were stained with Nissl bodies and had a layered sense.There was almost no abnormality in group A.The HE staining of hippocampal neurons in group S and Y+D+S was loose and the morphology was incomplete.Nissl-stained Nissl bodies are significantly reduced,and the layering is poor.Conclusion : 1.The spatial learning and memory ability of SD rats were significantly decreased after intermittent inhalation of sevoflurane,which proved that sevoflurane had an effect on learning and memory.2.Dexmedetomidine protects hippocampal neurons from SD rats induced by sevoflurane,and its mechanism may be achieved by α2-adrenergic receptors.
