| Objective: To study effect of Jia Jian Yi Jing Decoction on synapses development and plasticity in hippocampus of AD rats induced by ovariectomized.Methods: Female SD rats were randomly divided into Sham group,Ovx group,Ovx+L group,Ovx+M group,Ovx+H group and E2 V group,each group were ovariectomized expect the sham group.After 1 week of surgery,the treatment groups were gavaged with Jia Jian Yi Jing Decoction for 8 weeks,by the meantime,Sham group and Ovx group were gavaged with water.Tissue samples were taken after gavaged.Western Blot and Dot Blot were used to detect the expression levels of estrogen receptor,beta-amyloid protein,inflammatory factors,anti-apoptotic protein,synaptic protein and related protein kinase in hippocampus.The positive expression of NMDAR-2B and PSD95 in hippocampus was detected by Immunohistochemistry.Dendritic complexity and density of dendritic spines in hippocampus were detected by Golgi staining.Synaptic efficacy and plasticity were detected by Long-term potentiation.Results: After ovariectomized,the expression of ERα in rats was decreased,the loss of ERα could be reduced in all treatment groups.Jia Jian Yi Jing Decoction can increase the phosphorylation level of Akt,affect the activation of downstream GSK-3β,inhibit the deposition of Aβ,inhibit the pathology progress of AD,regulate the release of TNF-α,increase the expression level of anti-apoptotic protein Bcl-xL,improve learning and memory function.Golgi staining showed that Jia Jian Yi Jing Decoction could improve the complexity of dendrites and density of dendritic spines in hippocampus.Jia Jian Yi Jing Decoction could increase the expression of synaptic proteins,affect the activation of kinases,improve synaptic efficiency and plasticity,and then improve cognitive function.Conclusions: Jia Jian Yi Jing Decoction could improvability cognitive dysfunction in hippocampus of AD rats after ovariectomized.The mechanism was probably that it could ascension estrogen levels in brain,and activated Akt by increase the level of ERα,affected the activation of downstream GSK-3β kinase,exert the anti-apoptosis ability,induction of neurogenesis,increased the expression of synaptic protein and enhancing synaptic plasticity. |
