Objective:Prunella vulgaris L.,a genus of perennial herbaceous plants in the Labiatae family,is a commonly used traditional Chinese medicines.The chemical components are complex,the pharmacological actions are broad.At present,the main researches on Prunella vulgaris L.focus on its ears and the pharmacological activities of small molecule substances,including flavonoids,phenolic acids,triterpenoids,etc,but few are about its fruits and the macromolecular materials,such as Prunella vulgaris L.volatile oil.In this paper,components analysis of Prunella vulgaris L.fruits were researched,using Prunella vulgaris L.fruits as its raw material,and its antioxidant,anti-inflammatory activity are briefly discussed,which lay a foundation for the comprehensive utilization of Prunella vulgaris L.and the multi-direction development of Chinese medicinal materials.Method:1.Chemical components of volatile oil from 12 species of the Lamiaceae family plants fruits and 18 batches of Prunella vulgaris L.fruits were analyzed by GC-MS method,the common model and similarity of the TIC profiles were analyzed by Computer Aided Similarity Evaluation System,and the components of common characteristic peaks in chromatograms were identified.2.HPLC methods were applied to determination of fingerprints and main activecompounds contents of Prunella vulgaris L.fruits from different habitats.3.This paper studied the antioxidant ability of PSV and PSM in vitro by ABTSmethod,FRAP method,DPPH method.Inflammatory cell model was constructed by LPS acting RAW264.7 cell line,The concentration of IL-6,NO and TNF-αwere assayed by using ELISA,To evaluate the anti-inflammatory activity.Results:1.The main components of the volatile oil from 12 species of Labiatae fruits were unsaturated fatty acids.Among them,Prunella vulgaris L.contained Linoleic acid(39.32%),Squalene(4.63%)and Beta-amyrin(0.59%).2.GC-MS fingerprint of volatile oil from Prunella vulgaris L.fruit was established,and 19 common characteristic peaks were identified.3.The HPLC fingerprints of Prunella vulgaris L.fruits from different habitats were established,and the contents of isorosmarinic acid and isorosmarinic acid in the fruits were determined simultaneously.4.The results of anti-inflammatory bioactivity showed that the volatile oil and methanol extract from Prunella vulgaris L.fruits could significantly inhibit the LPS-stimulated RAW264.7 cells to secrete NO,TNF-αand IL-6(P<0.01)in a dose-dependent manner at the concentration of 23.493.8μg?mL-1.5.The antioxidant activities of volatile oil and methanol extracts from Prunellavulgaris L.fruits in vitro were studied.The results showed that DPPH,ABTS free radical scavenging activity and Fe2+reduction ability were good.Conclusion:In this paper,through GC-MS and HPLC analysis of Prunella vulgaris L.fruits,preliminary composition characterization confirmed that Prunella vulgaris L.fruits contain a large number of active ingredients,its volatile oil and methanol extracts have good anti-inflammatory and antioxidant effects.The established fingerprint provides an effective means for the quality control of Prunella vulgaris L.fruits,and provides an important basis for the comprehensive utilization of Prunella vulgaris L.and the multi-direction development of Chinese medicinal materials.
