Objective:To detect the expression of autophagy related gene LC3,tumor surface antigen NY-ESO-2 and MAGE-D4 and the infiltration of CD4+,CD8+,CD68+in colorectal cancer.To study the correlation among autophagy,immune cells and tumor surface antigen.Discuss the effect of autophagy on the immune function of colorectal cancer.Materials and MethodsThrough following up,The study group consisted of a total of 128 tissue samples and corresponding paraffin blocks with complete follow-up data were collected from the Department of Pathology of Medical University Hospital,China,from January 2010 to December 2011.All CRC patients were first discovered,and no patient had been treated with radiotherapy,chemotherapy or combined radiotherapy and chemotherapy before surgery.One-hundred-twenty-eight colorectal cancer cases were selected for immunohistochemical detection of CD4+,CD8+,CD68+ infiltration and the expressions of LC3,NY-ESO-2,MAGE-D4.Results1.Immunohistochemical results showed that the positive rate of LC3,CD4+、CD8+、CD68+ in colorectal cancer tissues was separately 67.19%,67.19%,48.44%,44.53%,47.66%,higher than those in normal mucosa group(P<0.05).Statistical analysis showed that the expression of LC3 was positively correlated with the infiltration of CD4+,CD8+,CD68+ cells.2.Immunohistochemical Western blots results showed that the positive rate of LC3,NY-ESO-2、MAGE-D4 in colorectal cancer was separately 67.19%,16.4%,61.72%,higher than those in normal mucosa group(P<0.05),western blots results showed that the expressions of LC3,NY-ESO-2,MAGE-D4 were higher than those in normal mucosa group.Statistical analysis showed that LC3 was positively correlated with the expressions of NY-ESO-2,MAGE-D4.3.Statistical analysis of the immunohistochemical results of NY-ESO-2、MAGE-D4 showed that the the expressions of NY-ESO-2 was positively correlated with the infiltration of CD8+,CD68+ cells,MAGE-D4 was positively correlated with the infiltration of CD4+,CD8+,CD68+ cells.4.The expression of LC3 and the infiltration of CD4+ and CD8+ immune cells are closely related to the site,staging and lymph node metastasis of the tumor.The expression of NY-ESO-2 and MAGE-D4 is closely related to staging and lymph node metastasis.Kaplan-Meier survival analysis showed that the expression of LC3 in colorectal cancer tissues,NY-ESO-2 and MAGE-D4 were positive,the 5 year survival rate with the tumor microenvironment in more CD4+,CD8+,CD68+ cell infiltration in patients was significantly higher than that of LC3,NY-ESO-2,MAGE-D4 and less negative expression of CD4+,CD8+,CD68+ cell infiltration in patients.Concluusion1.The autophagy in colorectal cancer was positively correlated with the infiltration of immune cells.The change of autophagy level in tumor cells may be an important factor affecting the immune function.2.The autophagy in colorectal cancer was positively correlated with the expressions of NY-ESO-2,MAGE-D4.The form of cell metabolism in autophagy can be one of the important factors to increase tumor surface antigen.3.Autophagy can change the nature of tumor surface antigen,increase the exposure of tumor surface antigen determinant,and induce specific tumor immunity and non-specific immunity,which may be a new way to induce tumor immune.4.Autophagy and its influence on cellular immune function in microenvironment are closely related to clinicopathological factors,it can affect the prognosis of colorectal cancer patients to some extent.
