Objective:To observe the expression of SIRT2 in HUVECs cell and to study the SIRT2 expression in HUVECs cell induced by ox-LDL.Methods:1.Culture of HUVECs cell:HUVECs cell were resuscitation,the cell were cultured in a premixed culture medium,and the changes of cell morphology and quantity were observed every day.2.Effects of different concentrations of ox-LDL on the viability of HUVECs cell by MTT assay.3.HUVECs cell were cultured in vitro and HUVECs cell were treated with ox-LDL to establish an inflammatory response model of endothelial cell.HUVECs cell was divided into three groups:control group(without ox-LDL),low concentration group(with ox-LDL 50 mg/L)and high concentration group(with ox-LDL 100 mg/L).HUVECs cell were treated with different concentrations of ox-LDL for 24h and 48h.4.HUVECs cell were cultured in vitro.The expression of SIRT2 and NLRP3 in HUVECs cell was detected by Western Blotting.5.HUVECs cell were cultured in vitro.The mRNA expression of SIRT2 and NLRP3 in HUVECs cell was detected by Real-Time PCR.Results:1.The expression of SIRT2 was found in HUVECs cell.2.Different concentrations of ox-LDL could inhibit the proliferation of HUVECs cell.3.With the increase of ox-LDL concentration,the expression of SIRT2 and NLRP3 increased gradually,and showed a concentration-dependent characteristic.Conclusion:ox-LDL can promote the expression of inflammatory factor NLRP3 in HUVECs cell and increase the expression level of SIRT2 at the same time.And both increased in a concentration-dependent manner.
