| Objective:To investigate the effect of Armillaria mellea polysaccharides on arterial thrombosis induced by ferric chloride with its mechanism.Methods:Forty SD male rats(body weight 200-250 g) were randomly divided into 5 groups.8 in each group,which were normal group,model group,aspirin group,low dose Armillaria mellea polysaccharides group,and high dose.Armillaria mellea polysaccharides group.The aspirin group was given aspirin 10 mg/kg by intragastric administration per day.The low-and high-dose Armillaria mellea polysaccharides groups of were given 100 mg/kg and 400 mg/kg of Armillaria mellea polysaccharides by intragastric administration per day.The normal group and the model group were given the same dose of normal saline by intragastric administration.After 7 days of intragastric administration,except for the normal group,the other models of the right common carotid arterv thrombosis were established with ferric chloride.After the modeling was completed,the common carotid artery at the thrombus formation site was cut off,the blood was collected from the heart,and the right brain tissue was taken from the broken head.Detection indices were as follows:(1)The weight of thrombus in each group of rats was weighed and calculated;(2)Flow cvtometry was used to detect whole blood platelet-leukocyte aggregates(PLA) levels in each group;(3)Plasma E-selectin,monocyte chemoattractant protein-1(MCP-1) and platelet-endothelial cell adhesion molecule-1(PECAM-1) level were detected by enzyme linked immunosorbent assay(ELISA).(4)Western Blot technique was used to detect the expression of Bcl-2 and Bax protein in the right brain tissue of each group of rats.Results:1.Changes in carotid artery thrombus weight in each group of rats:In the normal group,no blood vessel was weighed due to no thrombosis,and there was obvious intravasular thrombus formation in the model group.Compared with the model group,the weight of thrombus in the aspirin group was significantly reduced(P<0.01),the weight of thrombus in the low-and high-dose Armillaria mellea polysaccharides groups was reduced compared with the model group(P<0.05).2.Changes in platelet-leukocyte aggregate(PLA) levels in whole blood of rats in each group:Compared with the normal group,the PLA level of whole blood increased in the model group;compared with the model group,the PLA level in the high dose Armillaria mellea polysaccharides group decreased.3.Detection of levels of E-selectin.MCP-1 and PECAM-1 in plasma of rats in each group by ELISA:Compared with the normal group,the levels of E-selectin.MCP-1 and PEC AM-1 in the model group were significantly increased(P<0.01).Compared with the model group,the levels of E-selectin.MCP-1 and PEC AM-1 in the low-and high-dose Armillaria mellea polysaccharides groups were significantly decreased(P<0.01).4.Changes in the expression levels of Bcl-2 and Bax in the right cerebral tissue in each group of rats:Compared with the normal group,the Bcl-2/Bax ratio of the model group decreased significantly(P<0.01).Compared with the model group,the Bcl-2/Bax ratio of the low-and high-dose Armillaria mellea polysaccharides increased significantly(P<0.01).Conclusion:Armillaria mellea polysaccharides can inhibit the arterial thrombosis induced by ferric chloride,whose mechanism may be related to the reduction of vascular endothelial inflammation and the inhibition of platelet activation. |
