| Objective:To observe the effects of different acupoints on oxidative-antioxidant remodeling in rats with stress gastric ulcer(SU),and to provide the basis for optimizing acupoints for the intervention of SU.Methods: 50 male SD rats were selected and randomly divided into blank group,model group,positive drug group(ranitidine),acupuncture group 1(zhongwan and weishu),Acupuncture group 2(zhongwan and weishu)After feeding for 3d.Nine rats in each group.According to the method of bondage-water leaching mold making.The positive drug group was given ranitidine solution for gastric intervention,while the acupuncture group was given acupuncture intervention.After intervention treatment for 7 days,the model was built and executed.After tissue sampling,the gastric histopathological changes were observed by optical microscopy,and the contents of IL-6 and TNF-α in serum were detected by ELISA.Meanwhile,the contents of SOD and MDA in gastric tissue were detected.The expressions of TLR4,MyD88 and NF-ΚB in gastric tissues were detected by Westblot.Results:(1)observation results under light microscope of gastric tissues of SU rats in each group:Under the light microscope,Compared to the model group and the positive drug group.Acupuncture group 2 was more similar to that in the blank group.(2)results of serum IL-6 and TNF-α in serum of rats in each group:P < 0.05 was significant difference between the model group and the blank group.The positive group was significantly different from the blank group P < 0.05.Compared with the model group,both the acupuncture group and the model group had P < 0.05,and the statistical results were different,and the acupuncture group2 was better than the acupuncture group 1.(3)results of SOD and MDA examination in rat gastric tissues of each group: compared with the blank group,P < 0.05 was found in the model group.Compared with the model group,the expression of SOD increased and the expression of MDA decreased in the positive drug group.Compared with the model group,the increase of SOD and the decrease of MDA expression in the acupuncture group were significant.(4)detection results of TLR4,MyD88 and NF-ΚB in serum of blank group,modelgroup and hejiapaixue point group: TLR4 and other proteins were significantly expressed in the model group compared with the blank group.Compared with the model group,the serum TLR4 and other proteins in the acupuncture group 2were significantly reduced.Conclusion:(1)hemu point(zhongwan and zusanli)and sumu point(zhongwan and weishu)have positive effects on the recovery of oxidation-antioxidant system in the rat model of stress gastric ulcer,so as to prevent and treat stress gastric ulcer to a certain extent.(2)the repair effect of zhongwan and zusanli on oxidative damage in rat model of stress gastric ulcer is better than that of zhongwan and weishu.(3)zusanli and zhongwan may affect the oxidation-antioxidant system reconstruction of SU rats through TLR4/ NF-ΚB signaling pathway. |
