Objective:Y-maze and Morris Water Maze training tooks were chosen to establish the aninal model,to explore the association between learning/memory in mice(acquisition learning)and major histocompatibility complex polymorphism,the body’s defense ability.Try to attempt to further understand the factors that affect the learning/memory ability of mice from the perspective of molecular biology and the role of the mouse’acquisition learning in the body’s defense response.The learning process of mice can be dynamically observed by selecting the value of the slope of the latent period as an indication for the acquisition process.Using the characteristics of that advanced central nervous system of the body and the immune system play important role in the defense reaction,and all kinds of neural factors and inflammatory mediators affect each other,interact to resist foreign invasion,an infection model was established in the maze-trained mice to analyze the correlation between the advanced central nervous system and the immune system,so as to further understand the role of the advanced central nervous system in the body’s defense system.Methods:1.The differences in mice learning and memory ability in terms of experimental conditions and training time were analyzed,to establish a mouse learning/memory model suitable for this experiment,so as to provide a simple and reasonable training method for the experimental animal model.The experimental equipment of Y-maze was selected;mice were used to train by their nature of exploring novel behavior and enjoying dark environment.The experimental environment was set(experiment interval,training days),and the difference in learning and memory ability of mice was dynamically analyzed by the slope of the latent period as the judgment index.A slope<0 indicated learning during training and a slope>0 indicated no learning.2.Based on established training methods and judgment criteria for Y-maze,a model of learning/memory in experimental mice was established,The polymorphism loci were selected through the application of PCR(Polymerase Chain Reaction,PCR),agarose gel was used for imaging,and the relationship between the learning/memory ability of mice and the major histocompatibility complex was statistically analyzed.The learning and memory ability of mice was analyzed from the molecular perspective.A slope<0 indicated learning during training and a slope>0 indicated no learning.3.The experimental model of Morris Water Maze(MWM)was established according to the adaptability of mice to environmental changes.The slope of the latent period of the learning/memory ability in MWM was statistically analyzed to observe the correlation and difference with the results of Y-maze training.The model of infection was established in the mice after training,and the relationship between the learning/memory ability and the defense ability of body was preliminarily understood.Results:1.The slope of the latent period with an interval of 30s is better than that with an interval of 5s.From the beginning of training to the third day of training,the mice showed the best learning/memory ability,and the 3-day slope was selected as the indicator of statistical analysis.After the fourth day and the fifth day of training,their learning and memory ability did not make more significant progress,but the acquisition of learning reached a plateau after day3.2.Based on the 3-day slope,the two stains of mice were trained by Y-maze.The overall percentage of mice that learned was 46.1%.The percentage of mice that had learned with MudoEb5(37.9%)was significantly lower than that of mice without MudoEb5(61.1%;P<0.05).The percentage of mice that had learned with MudoEb7(26.1%)was significantly lower than that of mice without MudoEb7(51.9%;P<0.05).Differences between strains also led to different the learning/memory abilities in mice.3.By using the indicator of the latent period slope,the learning/memory ability of the mice in the MWM was analyzed,and it was found that the mice showed good performance on the third day after the MWM training.The correlation analysis between the survival days of mice and the 3-day slope showed a negative correlation.The survival days of mice with well learning/memory ability were longer,on the contrary,that the survival days were shorter.Conclusions:1.The major histocompatibility complex(MHC)and other alleles may be involved in the acquisition process.There may be a biological basis for learning in mice.H-2 class Ⅱ gene polymorphism was associated with learning and memory in mice,and others alleles may be involved in the acquisition process.The MHC may play an important role in animal cognition.2.The acquisition ability of mice was correlated with the immune ability of the body,the mice with better acquisition ability showed stronger performance in the process of anti-infection.The learning and memory ability of mice is related to the body’s defense ability to some extent.
