| Objective:To investigate the effect of sevoflurane postconditioning on mitochondrial autophagy mediated by Pink1/Parkin in myocardial ischemia/reperfusion injury.Methods:45 SD rats with a body mass of 200~250g at 10 weeks of age were randomly divided into Sham group(Sham group,n=15),ischemia/reperfusion injury group(I/R group,n=15),and ischemia/reperfusion injury sevoflurane postconditioning group(SPostC group,n=15).Myocardial ischemia/reperfusion injury model was prepared by ligating the left anterior descending branch of the coronary artery in rats for 30 min and reperfusion for 120 min.Intervention was performed in the SPostC group by inhaling 2% sevoflurane for 30 min after 120 min of reperfusion.Heart rate,blood pressure,heart rate and blood pressure product,myocardial infarction area,number of mitochondrial autophagosomes under electron microscope,and protein expression of key proteins LC-3 and Parkin in the Pink1/Parkin pathway were observed in each group.Results:myocardial infarction area of I/R group and SPostC group was larger than that of Sham group(P<0.05).Compared with the I/R group,the myocardial infarction area decreased in the SPostC group,and there was no significant difference in the number of mitochondrial autophagosomes under transmission electron microscope.There was no statistically significant difference in the expression of Pink1 and LC-3 proteins in the Pink1/Parkin pathway(P>0.05).Conclusion:sevoflurane postconditioning can reduce myocardial infarction area in myocardial ischemia/reperfusion injury in rats,but has no significant effect on mitochondrial autophagy in myocardial cells. |
