Objective: To investigate the expression and clinical significance of matrix metalloprotein 7(MMP7)and secretory crimp associated protein 5(SFRP5)in pancreatic ductal adenocarcinoma in Wnt signaling pathway.Methods: Firstly,the relative expression of MMP7 mRNA,SFRP5 mRNA in human normal pancreatic ductal epithelial cell line(HPDE6 cell line)and human pancreatic ductal adenocarcinoma cell line(PANC1 cell line)was detected by fluorescence quantitative PCR(RTFQ-PCR).Subsequently,45 cases of pancreatic ductal carcinoma and 45 cases of paracancerous tissues were collected from January 2017 to June 2018 in the Department of Hepatobiliary and Pancreatic surgery,affiliated Cancer Hospital of Xinjiang Medical University.To detect the expression of MMP7,SFRP5 in pancreatic ductal adenocarcinoma and paracancerous tissues,and to analyze the relationship between high expression and clinicopathological features,as well as its high expression in cancer tissues.The relationship between clinicopathological features and clinicopathological features.Results:.RTFQ-PCR showed that the relative expression level of MMP7 mRNA in PANC1 cell line was significantly higher than that in HPDE6 cell line,while the expression level of SFRP5 mRNA in PANC1 cell line was significantly lower than that in HPDE6 cell line(all P < 0.05).The high expression of MMP7 in pancreatic ductal adenocarcinoma was significantly higher than that in paracancerous tissues,while the high expression of SFRP5 was significantly lower than that in paracancerous tissues.The difference was statistically significant(all P < 0.05).Pearson chi-square test and Spearman grade correlation analysis showed that the high expression of MMP7 in cancer tissue was higher than that in pancreatic ductal carcinoma patients before operation,and the tumor size.There were significant differences in local lymph node metastasis,T stage,N stage and TNM stage,and there was a positive correlation between them.(all P < 0.05)The high expression of SFRP5 in cancer tissue was significantly different from local lymph node metastasis,N stage and TNM stage in patients with pancreatic ductal carcinoma(all P < 0.05).Logistic regression model multivariate risk analysis showed that the high expression of MMP7 in pancreatic ductal carcinoma was an independent risk factor for tumor size and local lymph node metastasis(all P < 0.05).The high expression of SFRP5 in cancer tissues was an independent risk factor for local lymph node metastasis and TNM stage in patients with pancreatic ductal carcinoma(all P < 0.05).There was a negative correlation between the high expression of MMP7 and SFRP5 in cancer tissues(r=-0.818,P< 0.01).Conclusion: Clinical monitoring of the expression of MMP7 and SFRP5 may provide a new candidate basis for early diagnosis of pancreatic cancer and effective therapeutic targets.
