Objective: To explore the protective effect of the Petroleum Ether Extract of Costusroot on ethanol-induced of gastric mucosa Injuries in rats.Methods: 1.Optimization of the extraction technology for the Petroleum Ether Extract of Costusroot With the extraction rate of Petroleum Ether Extract of Costusroot and content of costunolide and dehydrocostus as indexes,orthogonaltest was used to evaluate the effect of including extraction time,extraction temperature and solid-liquid ratio on extracting technology.2.The effect of Petroleum Ether Extract of Costusroot on gastric mucosa Injuries in rats The SD rats were divided into 6 groups.Group 1 wascontrol group,group 2 wasgastric mucosa injuries controls,another 3 groups each was administered with Teprenone,low-dose PEEC group(12.0mg/kg),middle-dose PEEC group(36.0mg/kg),high-dose PEEC group(108.0mg/kg)three groups treated with of Petroleum Ether Extract of Costusroot respectively.Group 1 and 2 were orally given 5% Tween80,once a day for five days in a row.On the sixth day,absolute ethanol(7.5m L/kg)was given to all the animals one hour later.All rats were monitored continuously for 1h thenwere sacrificed.Their stomachs were immediately removed,unfolded along the greater curvature and rinsed with cold saline,the area of ulcer was measured and inhibition percentage.Each gastric tissue was cut in half and prepared for histological studies.Hematoxylin-eosin and Periodic Acid Schiff reaction staining was performed to the gastric tissue to observe the pathological changes in the groups respectively.3.The effect and mechanism of Petroleum Ether Extract of Costusroot on gastric mucosa Injuries in rats The SD rats were divided into 6 groups.Group 1 was control group,group 2 was gastric mucosa injuries controls controls,another 3 group were administered with Teprenone,and low-dose PEEC group,middle-dose PEEC group,high-dose PEEC group(12.0mg/kg,36.0mg/kg,108.0mg/kg)three groups treated with of Petroleum Ether Extract of Costusroot.Respectively,Group 1 and 2 were orally given 5% Tween80 once a day for five days.On the sixth day,absolute ethanol(7.5m L/kg)was given to all the animals 1hlater.All rats were monitored continuously for one hour then were sacrificed.Their stomachs were immediately removed,unfolded along the greater curvature.Each gastric tissue was cut in three and were prepared to detecte the expression of MUC5 AC,TFF1 and TFF2 by immunohistochemistry,ELISA,and Western Blot methods.Results: 1.Optimization of the extraction technology for the Petroleum Ether Extract of Costusroot According to the extraction rate of Petroleum Ether Extract of Costusroot and content of costunolide and dehydrocostus as indexes,the results of orthogonal test showed that the optimum extraction time was 0.5h,extraction temperature was 60℃ and solid-liquid ratio was 1:10.2.The effect of Petroleum Ether Extract of Costusroot on gastric mucosa Injuries in rats Intragastric administration of the costusroot extracts protected gastric mucosa from ethanol-induced gastric ulcer reducing submucosal edema and preventing hemorrhagic damage.Meanwhile,it increased the production of gastric mucus.3.The effect and mechanism of Petroleum Ether Extract of Costusroot on gastric mucosa Injuries in rats(1)The expression of MUC5 AC,TFF1 and TFF2 were detected by immunohistochemical assay The signal for MUC5 AC,TFF1 or TFF2 protein in immunohistochemistry presenting brown color.Ethanol induced a marked decrease in the staining for MUC5 AC,TFF1 or TFF2 compared with the control group.In rats pretreated with Teprenone or Costusroots,the levels of MUC5 AC,TFF1 or TFF2 were upregulated compared with the ethanol ulcer group.(2)The content of MUC5 AC,TFF1 and TFF2 were detected by ELISA kits The content of MUC5 AC,TFF1and TFF2 was apparent decline compared to the normal control group(P<0.05,P<0.01).The rats pretreated with Teprenone,middle-dose PEEC and high-dose PEEC,the content of MUC5 AC,TFF1 and TFF2 were increased compared with the model group(P<0.05,P<0.01).(3)The protein expression of MUC5 AC,TFF1 and TFF2 was detected by Western Blot method The protein expression of MUC5 AC,TFF1 and TFF2 were down regulated in the model group when compared with the normal group(P<0.05,P<0.01).The rats pretreated with Teprenone,middle-dose PEEC and high-dose PEEC,the expression of MUC5 AC,TFF1and TFF2 was increased when compared with the model group(P<0.05,P<0.01).Conclusions: The Petroleum Ether Extract of Costusroot could reduce the area of gastric ulcer,alleviate the pathological damage of gastric tissue and increase the thickness of gastric mucus in model rats,thus effectively resist the injury of gastric mucosal induced by ethanol.Them echanism may be related to the upregulation of the expression of TFF1,TFF2 and MUC5 AC in gastric mucosa it enhanced the defensive function of gastric mucus barrier. |
