Objective: Observe the effect of Octreotide(OCT)on liver histopathology and liver serology in experimental hepatic fibrosis rats,and explore the role of RORα in octreotide-induced hepatic fibrosis in rats.Further revealing the specific mechanism of action of RORα against liver fibrosis,and providing new ideas for the treatment of liver fibrosis.Methods: Thirty-six male SD rats were randomly divided into three groups:control group,model group and OCT group.The model of hepatic fibrosis was established by CCl4(model group and OCT group).After 8 weeks of rearing,the rats were killed and randomly collected.The pathological changes of liver were observed by HE and VG staining.The levels of AST,ALT,HA,LN,PⅢNP and COL4.The degree of hepatic fibrosis and the degree of inflammatory activity in each group were analyzed by Ishak scoring system.The expression of RORα receptor in liver tissue was detected by Western blot.Results:1.Compared with the control group,the levels of hepatic pathological changes,inflammatory activity,fibrosis,serum AST,ALT,HA,LN,PⅢNP and COL4 in model group and OCT group were significantly increased and RORα significantly decreased P<0.01).2.Compared with the model group,the liver pathological changes,inflammatory activity,fibrosis,serum AST,ALT,HA,LN,PⅢNP and COL4 in OCT treatment group decreased significantly and the reduction of RORα decreased(P <0.05).3.Gross specimen changes: The color of the liver tissue of the control group was bright red,the liver edge was clear,not blunt,and the touch was extremely soft.In the model group,the surface nodules of the liver tissue changed greatly,the liver edge was round and blunt,the touch was hard,the color was light,and the fibrosis was clearly visible.The surface of the liver of OCT-treated rats was red with clear edges and soft touch.4.Pathological changes: In the control group,there were fewer inflammatory cells in the liver tissue,and apoptotic bodies were rarely seen.A very small amount of collagen fibers are distributed around the portal vein area,and the liver cells are usually arranged in a rope.The liver tissue of the model group was disordered,and the portal vein area and lobular fibrosis were obviously proliferated.Liver steatosis,flaky necrosis,mononuclear cell infiltration,lymphocytes and eosinophils;in the OCT group,hepatocytes were spherically swollen,with a large number of punctate necrosis,a small amount of debris necrosis,and a small amount of inflammatory cells infiltrating the liver tissue.Most of the cells are clearly arranged,and small collagen fibers surround the small blood vessels and portal veins.Conclusions: In rats,RORα decreased under the action of OCT,and it also protected liver function and reduced the degree of liver fibrosis.The therapeutic effect of experimental rat liver fibrosis was significant in this study..At the same time,this experiment is helpful to further reveal the specific mechanism of action of RORαagainst liver fibrosis,and also a new direction for the treatment of liver fibrosis.
