Aims of study:Transcription factor Sox2 is a member of the transcription factor SOX family,Sox2 plays an important role in the self-renewal and pluripotency of embryonic stem cells.Previous studies have found that Sox2 protein is highly expressed in the epithelium of the incisor,from the tooth plate epithelium of mouse tooth development E11.5 days to the labial cervical epithelium of the adult mouse incisor highly specific expression of Sox2 protein has always existed,However,whether the regulation of transcription factor Sox2 in the development of mouse incisor and the specific mechanism of action need to be further explained,The purposes of this study are as the followings:(1)Shh-cre,Sox2fl/fl conditional knockout mice were used to observe the phenotype of mouse incisor development;(2)To investigate the functional mechanism of transcription factor Sox2 in the development of mouse incisor.Material and methods:The construction of Shh-cre,Sox2fl/fl transgenic mice,The mutant and wild-type mice born at the same nest were chosen for the study,The experimental results of three different litters were used for the conclusion of this experiment.Through HE staining,immunohistochemistry,stereo microscope to observe the condition of the mouse incisor development phenotype,explore the influence mechanism of transcription factor Sox2 in mouse incisor development.Results:(1)The construction of Shh-creJSox2fl/fl transgenic mice,before the development of tooth germ(E10.5 days),Sox2 gene knockout in dental epithelium,which found that the mice died after birth,and the emergence of cleft palate,tongue and jaw adhesion phenotype.(2)The morphological observation of the embryonic tooth germ of mouse embryos showed that the Shh-cre,Sox2fl/fl transgenic mice had abnormal development of the incisor cervical loop,which could not form the typical structure of the incisor cervical loop.In addition,Shh-cre,Sox2fl/fl transgenic mice had abnormal appearance of the ameloblasts and odontoblasts.However,the effect of mouse molar on phenotypic development was not obvious.In the previously obtained K14-cre,Sox2fl/fl conditional knockout mice showed that loss of tooth germ phenotype in the mandibular third molar in the Shh-cre,Sox2fl/fl knockout mice did not appear.Conclusion:(1)The transcription factor Sox2 is also involved in maintaining the normal development of the palate and tongue(2)In the establishment of Shh-cre,Gox2fl/fl conditional knockout mice,mouse incisor tooth germ development affected,Sox2 gene can be used as a mouse tooth development is an essential transcription factor.
