Objective:To investigate the expression and clinical significance of proton pump(H+/K+-ATPase)in laryngeal carcinoma tissues.Methods:Selected from May 2013 to October 2016 in 30 patients with laryngeal carcinoma and 19 patients with benign laryngeal lesions after surgical treatment in the*****,Samples of paraffin blocks were extracted,including 30 laryngeal carcinoma tissue samples(laryngeal cancer group),17 adjacent laryngeal tissue samples(adjacent to cancer group)and 19 normal laryngeal tissue samples(normal control group).Immunohistochemical staining was performed to measure the levels of H+/K+-ATPase alpha and beta subunits.Statistical methods were used to analyze the different expression levels among the three groups,the relationships between the expression of H+/K+-ATPase alpha and beta subunits and the clinicopathologicai characteristics of laryngeal carcinoma patients,and the influence of H+/K +-ATPase alpha and beta subunits on disease-free survival rate of laryngeal cancer patients.Results:There were significant differences in the expression of H+/K+-ATPase alpha and beta subunits among the three groups(P<0.05).The positive rates of H+/K+-ATPase alpha and beta subunits in the laryngeal cancer group were 73.3%and 56.7%,respectively,which were significantly higher than those in the adjacent to cancer group and normal control group.The expression levels of H+/K+-ATPase alpha and beta subunits in laryngeal carcinoma tissues were not significantly correlated with sex,age,smoking history,drinking history,laryngeal cancer classification,T grade,clinical stage,pathological grade and lymph node metastasis(P>0.05).The 3-year disease-free survival rate of laryngeal carcinoma patients with positive expression of H+/K+-ATPase beta subunit was significantly lower than that of the patients with negative expression(P<0.05).The recurrence rate of H+/K +-ATPase alpha and beta subunit positive expression was higher than that of H+/K+ATPase alpha and beta subunit expression negative,but the difference between the two was not statistically significant(P>0.05).Conclusion:Proton pump(H+/K+-ATPase)alpha and beta subunits are highly expressed in laryngeal carcinoma tissues,but their expression levels are not closely related to the type,stage,pathological grade and lymph node metastasis of laryngeal carcinoma patients.Proton pump(H+/K+-ATPase)beta subunit may be of great value in prognosis evaluation of laryngeal carcinoma.
